In corneal transplantation,the receptor can identify the "non self" organs that enter the body and produce immune rejection.The rejection after transplantation is the main reason for the failure of corneal transplantation.Nowadays,the study of immune rejection has become an important direction of corneal transplantation.At present,the commonly used immunosuppressive drugs are not only expensive but also toxic and have adverse reactions to the body.The matrine from the legumes of Sophora flavescens roots,plants,fruits and other organic solvents extracted by ethanol,the acute rejection after transplantation to a certain extent,has the role of immunosuppression,while matrine cheap and small toxic and side effects of matrine drug use value get more and more study.In this study,matrine on rat immunological rejection after corneal transplantation operation to study the role of matrine as an immunosuppressive agent on CD4+CD25+regulatory T cells.In this study,Wistar rats as the research object,and the establishment of rat corneal transplantation model,by giving different concentrations of MT in rats after transplantation to study the experimental rat CD4+CD25+ regulatory T cells.The rats were randomly divided into control group and experimental group.After successful establishment of rat corneal transplantation model,the control group was given intraperitoneal injection of normal saline.The experimental group was injected with 5 mg / kg MT,10 mg / kg MT,15 mg / kg MT and 10 mg / kg of cyclosporin A for 7 consecutive days.According to the scoring criteria of corneal immune rejection,determine the growth of the graft after corneal transplantation.The peripheral blood and spleen of the rats in the control group and the experimental group were detected by flow cytometry 7 days after the operation.The peripheral blood of the rats in the control group and the experimental group was collected at 7d after the operation to carry out the enzyme linked immunosorbent assay(ELISA).Observed after transplantation graft can be found in the experimental group of corneal graft turbidity,degree of edema and neovascularization index lower than the control group.The result of flow cytometry showed that the content of CD4+CD25+ regulatory T cells in the 5mg/kg MT rats in the experimental group was higher than that in the control group on the 7th day after operation in peripheral blood and spleen.At the same time through the detection of rat peripheral blood can be found in the experimental group of IL-10 and TGF-?1 levels higher than the control group.The results showed that intraperitoneal injection of a certain concentration of matrine increased the content of CD4+CD25+ regulatory T cells and increased the levels of IL-10 and TGF-?1 in the cornea transplantation model.Certain concentrations of matrine can inhibit the immune rejection after transplantation.
