With the widely spreading of the notion of earlier diagnosis and therapy of breast cancer,the mortality of breast cancer had reduced by 25% during 1990 to 2007.Nevertheless,breast cancer is still one of the most common malignant tumor worldwide.And China contributed 12.2% of the number of globally newly diagnosed breast cancer.Besides,the morbidity of male breast cancer is showing a elevating trend in recent years.Although a couple of targeted drugs has been widely used in clinic therapies effectively,targeted drugs is not applied to any special types of breast cancer patients.There was no targeted drug against triple negative breast cancer(TNBC)so far.Being not sensitive to chemotherapy,the prognosis of this type of breast cancer not optimistic.Wilm's tumor 1 associated protein(WTAP)is a kind of protein binding to Wilm's tumor 1(WT1)gene specifically.WT1 is a gene inhibiting tumorigenesis and development of Wilm's tumor,and playing a vital role in the process of embryonic kidney and gonadal development.WTAP and WT1 mainly distributed in nucleus and partially co-located with splicing factor.WTAP had been detected in various of malignant tumor,such as breast cancer,colon cancer,bladder cancer and lung cancer,and worked as a key role in the development of embryo.In recent years,multiple of studies had demonstrated that WTAP worked in the process of tumorigenesis and development.WTAP had been proved that it promotes tumor proliferation and metastasis in many kinds of cancer,such as cholangiocarcinoma.However,the role of WTAP in breast cancer is still unclear.We will verify the function of WTAP in the process of proliferation,migration and invasion in this paper.Objective: To discover the influence of WTAP gene on the proliferation and migration and invasion activity of breast cancer cells.Methods:The Lentivirus of WTAP was used to transfect into breast cancer cell MDAMB-231.The WTAP m RNA and protein expression were accessed by Real Time-PCR and Western blot,respectly.Cell grow was measured by CCK-8.Matrigel-coated Transwell,Transwell inserts and wound healing had been used in examining cell migration and invasion vitality in vitro.Results: We successfully constructed the ovexpression and knockdown lentivirus of WTAP in MDA-MB-231,and termed as 231-WTAP and 231-sh WTAP,respectly.The expression level of WTAP m RNA and protein were notably reduced in 231-sh WTAP comparing with the negative control group(231-SCR),while increased in 231-WTAP,comparing with the negative control group(231-NC)(P<0.01).The capabilities of cell migration and invasion were weaker in group sh WTAP than those in group SCR,which were stronger in group WTAP than those cells in group NC(P<0.05).Conclusions: WTAP gene could remarkably promote proliferation,migration and invasion capabilities of breast cancer cell MDA-MB-231.we shall find the machanism of WTAP in breast cancer via further study,which will find out new directions for the clinical treatment of tripple negative breast cancer!...
