Objective To assess the feasibility and safety of temozolomide combined with etoposide for the treatment of adult high-grade gliomas.And to compare curative effects of temozolomide alone for postoperative patients with glioma.Summary the efficacy of two different chemotherapy regimens in the treatment of adult high-grade glioma.Methods We retrospectively analyzed 42 patients with adult high-grade glioma from June 2010 to June 2015 in our hospital.They were all treated surgically in our hospital with concurrent chemoradiotherapy.Then they were randomly divided into two groups,the single drug group(only temozolomide)and the united drug group(temozolomide combined with etoposide).The progressive free survival(PFS)and adverse effects were retrospectively observed.Kaplan-Meier survival analysis was used to compare the survival time and survival rate between the two groups.Log-rank test was used to compare the survival time.Chi-square test was used to count the data.The difference was statistically significant when the P<0.05.Results The single drug group included 22 patients took temozolomide alone,and the united drug group of 20 patients were treated with temozolomide combined with etoposide.There was no significant difference in gender,age,tumor grade,operation and so on between the patients of two groups.The median PFS and the PFS-6 months of the single drug group was 17.5 weeks and 31.8%.And,the median PFS and the PFS-6 months of the united drug group was 24.4 weeks and 45.0%.So the PFS and PFS-6 months of the united drug group is obviously higher than that of the single drug group(P<0.05).Meanwhile,the adverse reactions of patients were mainly gastrointestinal reactions(nausea and vomiting),transient liver and kidney function damage and myelosuppression.The number of the single drug group was 4,2 and 2.And the number of the united drug group was 4,3 and 2.The adverse effects between this two groups have no obviously differences(P>0.05).Conclusion In the postoperative comprehensive treatment of adult high grade glioma,the regimen of temozolomide combined with etoposide is feasible and safe.Compared with the single drug group,the median PFS and the PFS-6 months of the united drug group is better,with no remarkable increase in adverse reactions.
