Objective: To explore the effect of acupuncture on Glial scar and neuroinflammation.Methods: Ninety six Sprague-Dawley(SD)rats were randomly divided into normal group,model group and acupuncture group.And every group was divided into three-day(3d),seven-day(7d),fourteen-day(14d)and twenty-eight-day(28d)groups based on treatment duration,every group had eight rats.The moderate traumatic brain injury rat models was established by Feeney's free-fall epidural impact method.Acupuncture group was treated 15 minutes once a day after modeled in 24 hours.The chosen acupoints were Baihui(GV 20),Shuigou(GV 26),Fengfu(GV 16),Yamen(GV 15)and bilateral Hegu(LI 4).Model group did not accept treatment but fixed 15 minutes,normal group done nothing.Garcia JH method was used to evaluate neurobehavioral at first day after modeled and sampling day.The morphology of neurons and astrocytes were observed by transmission electron microscopy(TEM).Flow cytometry(FCM)was used to detect the percentage of astrocytes,neurons and microglia at 3d,7d,14 d and 28 d.Enzyme-Linked Immuno Sorbent Assay(Elisa)method was used to detect the inflammation factor expression of IL-6 and IFN-?.Results: 1.The Garcia JH neurobehavioral score results showed that the neurological score of model group was lower than normal group after modeled,the Garcia JH scores of acupuncture group were higher than model group in 3d,7d,14 d and 28d(P<0.05).2.The SEM results showed that the cell body of model group was swollen,chromatin dissolving and organelles were significantly reduced when compare with normal group.The cell deformability,chromatin slightly dissolving and organelles were reduced lightly in acupuncture group,no obvious abnormity in others.3.The FCM results showed that astrocytes and neurons in the model group were lower but microglia were higher than those in the normal group,the expression of astrocytes and microglia in acupuncture group were higher than model group at 3d but lower than model group at 7d,14 d and 28d(P<0.05),the expression of neurons in acupuncture group were higher than model group at 3d,7d,14 d and 28d(P<0.05).4.The results of Elisa showed that the inflammatory factors of IL-6 and IFN-? in model group were higher than those in normal group after modeled.The inflammation factor expression of IL-6 in acupuncture were higher than model group at 3d but lower than model group at 7d,14 d and 28d(P<0.05).The inflammation factor expression of IFN-? in acupuncture were higher than model group at 3d,7d,14 d and 28d(P<0.05).Conclusion: Acupuncture can regulate the repair of glial scar and the secretion of inflammatory factors in traumatic brain injury rats,and promote the recovery of nerve function after injury.
