Dragon's blood is the resin extracted from grease wood of liliaceous Dracaena cochinchinensis with alcohol.It possesses different pharmacological activities such as promoting blood circulation and arrest bleeding,antioxidant,antibacterial.It is extensively applied to heart disease,myocardial infarction and diabetes.To improve dragon's blood quality and achieve better therapeutically effect,this study investigates the extraction process of dragon's blood flavones using single factor experiment and orthogonal test,and the solid dispersion of total flavonoids of dragon's blood(TFDB)with a combination of Poloxamer 188(F68)and Polyvinylpyrrolidone k30(PVP k30).There is not a uniform method to determine the TFDB sinceits main componentsbelong to dihydrochalcone.In this study,the content of total flavonoids was determined by UV Spectrophotometry selecting LoureirinB as a standard and setting the detection wavelength at 277 nm.Standard curve equation was y = 50.3038 x – 0.0219 and Correlation coefficient r was 0.9995,suggesting that it wasin good liner relationship at the range of 1.24-12.4?g/ml.The recovery was 97~102%.These results indicated that such determinzation was stable,accurate and reliable satisfying the need of.of the measurement of TFDB.Reflux extraction with non-polarity solvent can effectively enhance the content of TFDB.Single factor test and orthogonal test were employed to optimize the extraction process of total flavonoids from dragon's blood.The optimal extraction technology was as follow: 10 times amount of ethyl acetate and once extraction for 0.5 h at 70?.This process produced the total flavonoids with an content of 54.61%,which meet the requirement of the fifth traditional Chinese medicine for the extraction of effective parts of plant medicine in drug registration regulation.Preformulation study was conducted to get some characteristics of TFDB.The equilibrium solubility of TFDB in water is 3.88 ?g/ml,whereas the equilibrium solubilities in the buffer salts are greater more,it also showed a trend that equilibrium solubility increased with the increased pH value.Factors tests showed that in the light,humidity and 40? condition was stable but while decreased in the 60?.Research on the materials showed that the use of the carrier material does not interfere with the determination of total flavonoids in Dragon's Blood.Solid dispersions are prepared by using F68 and PVP k30,and fitting a dissolution curve of the better one.F68 and PVP k30 can improve solubility and dissolution rate of TFDB.The analysis showed that the solid dispersion prepared with TFDB: PVP k30: F68 at mass ratio of 1: 8: 1 had a higher solubility and faster dissolution rate than its physical mixture,and the resulting dissolution curve matched the Weibull model.The solid dispersion was characterized by a combination of X-ray diffraction(XRD),differential scanning calorimetry(DSC),infrared spectroscopy(IR)and scanning electron microscopy(SEM).The phase identification for solid dispersion shows that the drug particles decreased,the hydrogen bond produced in drug and carrier,and novel amorphous status formed by using...
