Objective:To establish a new rat model of coronary microembolization(CME)by injecting autologous micro thrombus particles via right common carotid artery with cardiac catheterization.And to appraise the effects of Tongluo decoction on the coronary microembolization in rats.Methods:1.To establish rats model with coronary microembolization:The 36 male Sprague-Dawley(SD)rats were randomly divided into 4 groups:24 hours model group coronary microembolization(n=10),30 days model group coronary microembolization(n=10),sham group(n=10)and normal group(n=6).Coronary microembolization in rats were established by injecting0.3-0.5ml suspension of autologous micro thrombus particles into the root of aorta with ballon catheter through the right common carotid artery.2.Intervention of Tongluo decoction on the coronary microembolization in rats:26 male SD rats were divided into treatment group(n=10),control group(n=6),normal group(n=10).Hematoxylin-eosin(HE)staining and martius-sarlet-blue(MSB)staining were used to observe the number of positive arteries embolization.Hematoxylin-basic-fuchsin-picric(HBFP)staining was used to study the area of myocardial ischemia.The level of serum cTnI was tested by ELISA.The expression of TNF-? and IL-1 ? in myocardium were tested through immunohistochemical staining.Results:1.The numbers of positive arteries embolization of the 24 h model group and 30 d model group were significantly higher than that of the sham group(P<0.05)in the HE staining and the MSB staining.The areas of myocardial ischemia in the 24 h model group and 30 d model group were significantly higher than that of the sham group in the HBFP staining(P<0.05).Compared with sham group,the level of serum cTnI of model group was obviously increased(P<0.05).The expression of TNF-? and IL-1? in myocardium of the 24 h model group and 30 d model group were obviously higher than that of sham group(P<0.05).2.After intervention of Tongluo decoction,the embolus of coronary microembolization of Tongluo decoction group was becoming smaller than that of the control group and obviously organized in HE staining.In the myocardium HBFPstaining,the positive staining area of Tongluo decoction group was significantly lower than that of the control group(P<0.05).Compared with Tongluo decoction group,the level of serum cTn I of the Tongluo decoction group decreased(P<0.05).The expression of TNF-? and IL-1? in myocardium of the Tongluo decoction group was obviously lower,compared with the control group(P<0.05).Conclusion:1.The rat model of coronary microembolization can be built successfully and economically by injecting autologous micro thrombus particles into the root of aorta via the right common carotid artery with ballon catheter.2.The Tongluo decoction could promote the recanalization of arteriole microemboli,improve blood flow of embolism arteriole,and inhibit the inflammatory response in myocardium of CME rats.Tongluo decoction can reduce the ingury of coronary microembolization to the myocardium of rats.
