Objective:We aim to investigate the expression of PLAC1 in pancreatic cancer and its clinical association in pancreatic cancer patients,in order to provide a new prognostic biomarker and therapeutic target for pancreatic adenocarcinomaMethods:93 samples were collected from PC patients who underwent surgical resection at Fuzhou Dongfang Hospital of Xiamen University and Xijing Hospital of fourth Military Medical University during 2004 to 2015.Formalin-fixed paraffin-embedded cancer and its non-cancer tissue counterpart were used.Follow-up was performed for all the patients through telephone call and/or outpatient clinic.Immunohistochemistry was performed to evaluate expression of PLAC1.PLAC1 expression of all samples was scored by the semi-quantitative scoring method.Analyses of statistics were done with SPSS 19.0 software.To evaluate the correlations between PLAC1 expression and patients' clinicopathologic characteristics,Fisher's exact text and ?2 test were applied.Kaplan-Meier curves were depicted to reflect the overall survival and the differences between the 2 curves were analyzed by log-rank test.Univariate and multivariate Cox regression analyses were performed to assess the prognostic significance of PLAC1 expression.Results:1.The expression of PL AC 1 was detected in 41(44.1%)patients,whereas no staining was present in adjacent non-cancer tissues.2.Among patients' clinicopathologic characteristics,PLAC1 expression was only significantly correlated with tumor differentiation(P=0.028).3.Univariate analysis revealed that PLAC1 expression(p=0.016)and tumor differentiation(p=0.003)and were significantly correlated with poor survival in the whole cohort.4.Subgroup analysis showed PLAC1 expression was an independent prognostic biomarker in the perineural invasion positive subgroup(HR=2.950,95%CI 1.322?6.580,P=0.008).Conclusions:The protein expression of PLAC1 associated significantly with decreased overall survival in patients with PDAC,indicating that it was a valuable prognostic marker for PD AC and might be a potential target for immunotherapy.
