We have known that the transcription factor AP-2? serves as a tumor suppressor.Tumor suppressors are commonly methylated in gliomas.We analyze whether AP-2? has been methylated and acetylated in gliomas.It will be valuable for identifying signal pathways of gliomas and finding other treatment strategy.Firstly,we performed the IHC to analyze the expression levels of AP-2? in glioma and found that AP-2? is lowly expressed in high-grade glioma.However,adjacent normal tissues showed high AP-2? expression.And western blots revealed median AP-2? expression in U251 compared to SHG-44 with relatively low expression.So we traeted the genomic DNA of both cells with sodium bisulfite and carried out PCR amplication using methylation and ummethylation primers and identified AP-2? to be methylated in gliomas.Moreover,reexpression of AP-2? restrains the growth of gliomas treated with DNA methyltrasferases inhibitors and histone deacetylase inhibitors by western blot and MTT assays.We found the expression of AP-2? is increased and gliomas are inhibited.In conclusion,these data demonstrated that AP-2? is methylated to affect glioma celluar growth.
