| This paper focuses on the research of RP-HPLC chromatogram of Angelica Dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.var.?ormosana(Boiss.)Shan et Yuan,Then the metabolic prediction technology combined with virtual screening method and pharmacodynamic experiment,Screening the pharmacodynamics substances for migraine treatment.finally,The Real-time Quantitative PCR was used to test the inhibitory effect of imperation to the mRNA of migraine related target genes NF-κB,CGRP,TNF-α,iNOS,eNOS,c-fos and c-jun.In order to further study the relief effect and mechanism of active ingredient of Angelicae Dahuricae in nitroglycerin-induced rat migraine.In the research of RP-HPLC chromatogram of Angelica Dahurica,The RP-HPLC chromatogram of the 10 batches of Angelica Dahurica and similarity evaluation is established with good reproducibility and stability,And prove the 10 batches of samples similarity is higher,the chromatogram can fully control the quality of the Angelica Dahurica.In the study of efficacy material based on virtual screening,Twenty chemical components in Angelica Dahurica was docked with eight target related to migraine by using the Molegro software,and the effects of Angelica Dahurica in nitroglycerin-induced rat migraine were studied.The experimental results show that byakangelicin,scopolin,oxypeucedanin hydrate,imperatorin have an significant effect to migraine models.Research on mechanism of imperation in nitroglycerin-induced rat migraine.The qPCR assays was used to test the effect of imperation to the mRNA of migraine related target genes NF-κB,CGRP,TNF-α,iNOS,eNOS,c-fos and c-jun.The experimental results show that the high dose group of imperation can significantly reduce the mRNA expression levels.Therefore,imperation can relieve and treat the nitroglycerin-induced rat migraine by inhibiting the inflammatory factors,regulating the vascular tone and blood flow distribution,as well as reducing or blocking the transduction of pain stimulation in the central nervous system. |
