Effect Of Yiqi Yangyin Quyu Chinese Herb On TLR-IFN-BAFF Signaling Pathway In Patients With Primary Sj?gren's Syndrome

Objective To investigate the effect of Yiqi Yangyin Quyu traditional Chinese herb on the expression of TLR9,IFN-a and BAFF in serum of patients with Sj?gren's syndrome,and to investigate the effect of Yiqi Yangyin Quyu Chinese herb on TLR-IFN-BAFF Signaling Pathway in patients with Sj?gren's syndrome.Method 60 patients with primary Sj?gren's syndrome were randomly divided into two groups:the TCM group(treat with Yiqi Yangyin Quyu traditional Chinese herb)and the western medicine group(treated with hydroxychloroquine),each group with 30 patients.20 Healthy subjects as a normal control group.Clinical symptoms(mouth parched VAS score,Dryness of eye VAS score,Joint pain VAS score and SSDAI score)and blood index(erythrocyte sedimentation rate,serum IgG content)were observed before and after treatment in TCM group and western medicine group.Through enzyme-linked immunosorbent assay measure the levels of TLR9,IFN-a and BAFF protein in 60 Sj?gren's syndrome patients before and after treatment and 20 healthy subjects.Results 1.There was no significant difference in age,sex and weight between the TCM group,the western medicine group and the healthy control group(P>0.05).The course of the TCM group and the western medicine group before treatment,There were no significant differences in mouth dry VAS score,eye dry VAS score,joint pain VAS,SSDAI score,ESR level and serum IgG level between the two groups(P>0.05).2.After treatment,the level of ESR in the TCM group was significantly lower than that before treatment(P<0.01).The level of serum IgG in the TCM group was lower than that before treatment(P<0.05).the level of ESR and serum IgG in the western medicine group was significantly lower than that before treatment(P<0.01).3.After treatment,the TCM group and the western medicine group had a statistically significant differencein the clinical symptoms such as dry mouth VAS score,eye dry VAS score,joint pain VAS score and SSDAI score compared with before treatment(P<0.01).The VAS score of the joint pain of the western medicine group after treatment was lower than that of the traditional Chinese medicine group,and the difference was statistically significant(P<0.05).The VAS score of dry mouth and the VAS score of the dry eye of TCM group after treatment was lower than the western medicine group after treatment(P<0.05).4.The levels of serum TLR9 protein,IFNa protein and BAFF protein in the TCM group and the western medicine group were higher than those in the healthy control group before treatment,the difference was statistically significant(P<0.01).After treatment,the levels of serum TLR9 protein,IFNa protein and BAFF protein in TCM group and western medicine group were significantly lower than those before treatment(P<0.01).Conclusion 1.Yiqi Yangyin Quyu traditional Chinese herb can improve the clinical symptoms of patients with Sj?gren's syndrome,and significantly improve the mouth parched and Dryness of eye,while reducing ESR and serum IgG content of patients with Sj?gren's syndrome;2.The levels of TLR9,IFN-a and BAFF in serum of patients with Sj?gren's syndrome were higher than those of normal subjects,it suggested that TLR-IFN-BAFF Signaling Pathway play an important role in Sj?gren's syndrome;3.The expression of TLR9,IFN-a and BAFF protein in serum of patients with Sj?gren's syndrome reduced by treat with Yiqi Yangyin Quyu Chinese herb.This suggests that the Yiqi Yangyin Quyu Chinese herb may be caused by the effect of TLR-IFN-BAFF signaling pathway to play a therapeutic role with Sj?gren's syndrome.
