Chapter one,Literature study.A synthetical review was given to the ancient and present research of Danggui-Chishao drug pair from home and abroad.Chapter two,the study of the characteristics of the component dissolution in vitro of Danggui-Chishao drug pair.Multiple techniques of UPLC-Q-TOF/MS and UPLC-TQ/MS were applied to analyze the characteristics of the component dissolution in vitro of Danggui-Chishao drug pair in different extractions and anterior-posterior compatibility.It was demonstrated that most dissolution rates of the bio-active components were higher in water and low concentration alcohol and compatibility could change the dissolution rate while rates of components in Danggui decreased with the increasing of Chishao ratio,and rates of components in Chishao decreased with the increasing of Danggui ratio,ratio from 1:3 to 3:1 had better.dissolution condition.Chapter three,the study of the characteristics of efficacy preference of the compatibility of Danggui-Chishao drug pair.Section one,the data mining study of the characteristics of efficacy preference of the compatibility Danggui series drug pairs.Data mining method was applied to analyze the prescriptions containing Danggui series drug pairs in "lexicon of TCM prescription".Results displayed that the main efficacy was blood activing and nutriting(the number was more than 50%of the totle).There were higher frequency of some prescriptions like Danggui-Baishao,Danggui-Huangqi,Danggui-Shudi in nutriting blood than others,offering these prescriptions prefer to nutriting while others disaccord.Section two,the pharmacology experiment study of the characteristics of efficacy preference of the compatibility Danggui series drug pairs.The blood-deficiency mouse model induced by cyclophosphamide and acetylphenyhydrazine was applied to analyze the blood nutriting efficacy of Danggui-Chishao drug pair.Periphera hemogram,immune organ index and BRC ATPase activity were made statistics based on PCA,the scores of Danggui-Baishao,Danggui-Huangqi,Danggui-Shudi were more closer to normal group,offering these drug pairs had better blood nutriting efficacy.Section three,the compatibility study of the blood activing efficacy of Danggui-Chishao drug pair.The acute blood stasis rat model induced by subcutaneous injection of epinephrine hydrochlaride and ice-water stimulation was applied to analyze the blood activing efficacy of Danggui-Chishao drug pair.Haemorheology,blood clotting tetrachoric index were made statistics based on PCA,the scores of the dosed group whose ratio from 1:3 to 3:1 were closer to normal group,offering these group had better blood activing efficacy than other ratioes and single drug.Chapter four,the study of the characteristics of the metabolism of Danggui-Chishao drug pair.Section one,the study of the metabolites of normal rat after intragastric administration of Danggui-Chishao drug pair.UPLC-Q-TOF/MS and MetaboLynx software based on mass defect filter(MDF)were applied to analyze the metabolites in the urine and plasma of normal rats after intragastric administration of Danggui-Chishao drug pair.Seven metabolites of ferulic acid such as ferulic acid sulfate,cis-ferulic acid,ferulic glucuronate and so on;seven metabolites of gallic acid such as methoxyl hydroxybenzoic acid,hydroxybenzoic acid and so on;the metabolites of ligustilide and butylphthalide were identified as riligustilide and hydroxybutylphthalide respectively.Section two,the study of the endogenous metabolism of acute blood status rat after intragastric administration of Danggui-Chishao drug pair.UPLC-Q-TOF/MS and MarkerLynx software based on multivariate statistics were applied to analyze the influence on the endogenous metabolism of acute blood status rat after intragastric administration of Danggui-Chishao drug pair.Eight potential bio-markers such as kynurenic acid,lysophosphatidic choline,allantoin and so on were identified consequently,infering it effected based on the multitarget like HPA sympathetic nervous system and RAAS,referring lipoidic metabolism and some amino acid metabolism.
