Objective:To study The effect of migration and invasion research of BAPN on human osteosarcoma MG-63 cells and further explore the role of gene Lox?Mmp-2 and Mmp-9 in it.Methods:(1)Human osteosarcoma Mg-63 cells were cultured at logarithmic phase,the stduy was divided into control group and different concentration groups of drug,the morphologic changes of cells were observed by inverted phase contrast microscope..(2)Detection of proliferation rate of Mg-63 cells by CCK-8 assay.(3)detection of invasion and migration abilities of Mg-63 cells by Transwell assay(4)Expression level of gene LOX,and migration related gene MMP-2?MMP-9mRNA were measured by RT-PCR.(5)Expression level of LOX?MMP-2?MMP-9protein were measured by Western Blot.Results:(1)the morphologic changes of cells were observed by inverted phase contrast microscope,the mg-63 cells were divided into the control group and three different concentration groups of drug(2)CCK-8 assay test showed that proliferation of cells of drug groups was inhibited,The drug resistance index(IC50)was 0.65mM?0.32mM?0.19 mM,(P <0.05).(3)Transwell assay test showed that drug groups had higher invasion and migration abilities compared with control group,and there was statistical significance between drug groups and control group(P<0.05).(4)The results of RT-PCR showed that compared with comtrol group,expression level of migration related gene MMP-2?MMP-9 of drug groups were increased gradually(P<0.05),while gene LOX was decreased gradually(P<0.05).(5)The results of Western Blot showed that compared with comtrol group,expression level of MMP-2?MMP-9 protein were increased gradually(P<0.05),while LOX protein was decreased gradually(P<0.05).Conclusion:(1)When LOX was inhibited by BAPN,it was able to increase the human osteosarcoma Mg-63 cell migration and invasion ability.(2)The suppressed LOX was able to increase the human osteosarcoma Mg-63 cell migration and invasion ability by up-regulating express level of gene MMP-2 and MMP-9.
