Effect of echinococcosis granulosa on the distribution of immune cell subsetsObjective: Study through the detection of echinococcosis granulosa in patients with cystic fluid in vitro and its influence on the distribution of subtypes of immune cells,understand the hydatid disease for the body immune cell subsets will produce what effect.Methods:1.Collected 32 cases of patients with hydatid disease from January to July in 2016,during the cell transplantation center of Sichuan Provincial People’s Hospital for treatment of peripheral blood,according to whether or not to accept the surgical resection of the lesions could be divided into before surgery treatment group(n=16)and after surgery treatment group(n=16).Get after treatment in whole blood of peripheral blood mononuclear cells by adding corresponding monoclonal antibodies of the immune cells to investigate distribution of NK cells,CD4+T cells and CD8+T cells in peripheral blood mononuclear cell.2.Hydatid cyst fluid from patients in the surgery of hydatid cyst resection,centrifuge and filter bacteria processing of cystic fluid in vesicles which is obtained and used in the expriment.Spleen cells that are from 8~10 week female C57/B mice and hydatid cyst fluid mixed cultivation,in the experimental group,the spleen cells of mice were mixed with the protein level of 5mg/ml,1mg/ml and 100ug/ml,respectively,the control group was fed with complete medium and mouse spleen cell culture.The distribution of CD4+T cells and CD8+T cells in mouse spleen cells were detected by flow cytometry after mixed culture of 24 h and 48 h.Result1.The distribution of NK cells,CD4+T cells,CD8+T cells and CD4/CD8 in whole blood mononuclear cells(PBMC)before and after surgery was not statistically significant.(P>0.05)2.The distribution of effector CD4+T cells before and after operation in patients with whole blood mononuclear cells was statistically significant(P<0.05),There was no significant difference in the distribution of effector CD8+T cells before and after operation.(P>0.05)3.After the mice spleen cells were cultrued with the cyst fluid,it was found that the inhibitory effect of the cystic fluid on the distribution of CD4+T cells was statistically significant(P<0.05).However,the effect of cyst fluid on the distribution of CD8+T cells was not statistically significant.(P>0.05)Conclusion:1.In patients with hydatid lesions were resected,with effect of peripheral blood CD4+T cell percentage of immune cells in the group increased.2.Hydatid cyst fluid had inhibitory effects on the distribution of CD4+T cells in mouse spleen cells.To explore the effect of hydatid cyst fluid for immune cell energy metabolismObjective: To investigate the existence of hydatid cyst fluid reprogramme on immune cell energy metabolism phenotype,to provide new ideas for the study on the immunity escape mechanism of hydatid disease.Methods: Hydatid cyst fluid from patients in the surgery of hydatid cyst resection,centrifuge and filter bacteria processing of cystic fluid in vesicles which is obtained and used in the expriment.Jurkat cell lines from liquid nitrogen to recovery,the recovery after generation once passaged Jurkat cells and hydatid cyst fluid mixed culture 24 h as the experimental group(n=10),the Jurkat cells that did not add the cystic fluid were control group(n=10).The Cell energy phenotype distribution of Jurkat cells was detected by Extracelluar Flux AnalyzerResult:1.Jurkat cells and hydatid cyst fluid mixed culture group in energy metabolism potential was significantly lower than the control group.2.In the experimental group,the energy metabolism phenotype of Jurkat cells after the addition of drug stimulation was in the glycolytic quadrant,In the control group,the energy metabolism phenotype of Jurkat cells was in the Energetic quadrant.Conclusion:1.The potential ability of hydatid cyst fluid on energy metabolism of Jurkat cells was inhibited.By inhibiting the energy metabolism potential of immune cells,the biological function of immune cells is affected and the immune response is suppressed. |
