Objective: To clarify whether the expression of gastrokine 1(GKN1)help to estimate the risk classification in the gastric GIST,and then guide the treatment and evaluate prognosis,we detected the expression of GKN1 in gastric GIST and peritumoral tissue and analyzed the relationship between the expression level of GKN1 and biological behavior of gastric GIST.Methods:1.Gastric GIST and adjacent normal tissues of 30 gastric GIST patients were selected from the paraffin specimens in the department of pathology.The expression values of GKN1 in these tissues were detected by immunohistochemistry.2.The expression of GKN1 in 20 cases Fresh specimens of gastric GIST and adjacent normal tissues was quantified by using real-time fluorescence quantitative polymerase chain reaction(RT-PCR)and Western Blot.Meanwhile,the informations consisted of sex,age,tumor size,mitotic index,risk assessment and other information of the patients were collected.Finally,the experimental data combined with clinicopathological informations were analysed.Results:1.Immunohistochemistry showed GKN1 mainly expressed in the gastric mucosa,and little or no expression in gastric GIST tissue.the expression of GKN1 in 30 patients,adjacent normal tissues> gastric GIST tissue: 26 cases,adjacent normal tissues <gastric GIST tissue: 1 case,adjacent normal tissues = gastric GIST tissue: 3 cases,the expression of GKN1 in the gastric GIST was significantly lower than in peritumoral tissue.2.RT-PCR and Western blot showed that the expression GKN1 in gastric GIST tumors was significantly lower than in adjacent tissues(p<0.05),Meanwhile western blot revealed that GKN1 highly expressed in a very low risk of gastric GIST,and was Lower expression in in other risk of gastric GIST.3.There was no significant relationship between the expression of GKN1 gene and the sex,age,mitotic index of gastric GIST.The expression of GKN1 mRNA in diameter(?5cm)of gastric GIST was significantly higher than the diameter(> 5cm)group,and in Very low risk/low risk group was significantly higher than moderate/high risk group(p < 0.05)Conclusion:1.GKN1 mainly expressed in the gastric mucosa layer,the expression of GKN1 in gastric GIST is significantly lower than in adjacent tissues.2.There is no relationship between the expression of GKN1 and the mitotic index of gastric GIST,but the expression of GKN1 has certain relevance with the diameter of gastric GIST,when tumor diameter is greater than 5 cm,GKNl expression is obviously decreased.3.The expression of GKN1 in very low risk/low risk group is significantly higher than moderate/high risk group,this show that GKN1 is decreased from very low risk level to high risk level in gastric GIST,and it has some clinical reference value in identifying GIST biological behavior.
