EZH2,E-Cadherin In The Expression And Significance Of SD Rats With Endometriosis

Objectives:The aim of this study was to discuss the growth of ectopic lesions and the expression of EZH2 and E-cadherin in the patients with endometriosis of SD in the pr esence of EZH2 inhibitor(3-Deazaneplanocin A,DZNep)and to analyze the relations hip between them,in order to discuss the role of them in the pathogenesis of endometr iosis disease,to provide some test reference for the long-term clinical diagnosis and dr ug treatment.Methods:The rat model of endometriosis by autologous transplantation was establi-shed.After four weeks of transplantation,succefful establishment of the model was verified by laparotomy.Following the measurement of the volume by the vernier caliper,the rats were divided into three groups randomly:Experimental Group(treated with 3-Deazaneplanocin A,injection 2mg/kg twice a week for 2 weeks,n=16),PBS Group(treated with equal volum of PBS buffer,n =16),Control Group(without treatment,n =15)and Blank Group(health rats without laparotomy,n= 15).The volum of ectopic foci were measured after two weeks.The expression of EZH2 and E-cadherin were examined by Western Blotting and immunohistochemistry(IHC).Results:?Of all 65 rats,47 cases were succeeded to establish the model by the autologous transplantation method,the achievement ratio being 72.30%(47/65×100%).?The lesion volume after treatment in experimental group was decreased compared to that before treatmentm with a significant difference(53.21± 11.46 mm3 v.s.8.79±2.79mm3,P<0.05).?The expression of EZH2 were statistically significant decreased in experimental group after medication(P<0.05).By the immunohistochemical detection,the mean positive expression density of EZH2 protein in ectopic foci tissue was 35.30± 1.64(experimental group),41.70± 1.38(PBS group),43.10± 1.52(control group),32.70± 1.64(endometrial tissue of blank group).The mean positive expression density of E-cadherin protein in ectopic foci tissue was 21.20±1.64(experimental group),13.92±1.48(PBS group),15.70±1.64(control group),23.30±1.64(endometrial tissue of blank group).The relative expression of EZH2 protein of ectopic foci tissue by Western Blot was 120.76(experimental group),185.21(PBS group),195.26(control group),85.74(endometrial tissue of blank group).Both differences were statisticlly significant.? After intervention,the expression of EZH2 protein of ectopic foci tissue in experimental group was significantly lower than that in PBS group and control group,the expression of E-cadherin protein of ectopic foci tissue in experimental group was significantly higher that in PBS group and control group.Conclusions:(1)There was a high expression of EZH2 and low expression of E-Cadherin in th e endometriosis of SD rats.(2)EZH2 inhibitor(DZNep)can reduce the volume of ectopic lesion in SD rats,which is related to the high expression of EZH2 and the low expression of E-cadherin in endometriosis.(3)High expression of EZH2 and low expression of E-Cadherin may be associate d with distant metastasis and invasion of endometriosis.(4)The overexpression of EZH2 and low expression of E-cadherin in endometrio sis,not only explained one of the pathogenesis of endometriosis,but also provided ex perimental basis for the future development of new drugs.
