Objective:The aim of this study was to investigate the expression of amphiregulin(AR)in hepatocellular carcinoma(HCC),and exploring the relationship between the expression of AR and the clinicopathological parameters,pathologic differentiation and prognosis of HCC.For provide some experimental data for the search of new liver cancer diagnostic index and potential therapeutic target.Method:1.In this study,we selected 110 patients with radical resection of HCC from May 2009 to December 2011,and the pathological specimens inclouding HCC tissues and adjacent normal liver tissues(ANLT).2.We used Real-time PCR method to detect the expression of AR mRNA in all pathological specimens.3.We used immunohistochemical method and western-blot to detect the expression of AR protein n all pathological specimens.Result:1.There was no significant correlation between the expression of AR and the clinicopathological parameters of patients with hepatocellular carcinoma(HCC).There was no significant correlation between AR expression and age,AFP,lymph node metastasis,tumor diameter and tumor staging(P> 0.05)The degree of differentiation and the number of tumors were correlated(P <0.05).2.AR expression level and the degree of differentiation,the total survival time is closely related.The survival time of patients with high AR expression was significantly higher than that of AR patients(P <0.05).The expression of AR in the differentiated tissues was significantly different from that in the differentiated tissues.The expression of AR was significantly higher in the well differentiated and poorly differentiated hepatocellular carcinoma tissues than in the adjacent tissues.Lower than the adjacent tissue.3.The results of RT-PCR detection of AR mRNA in HCC showed that AR mRNA was significantly higher in patients with well differentiated hepatocellular carcinoma than in adjacent tissues,while in AR tissues and tissues of patients with moderate to differentiated hepatocellular carcinoma,Lower than the adjacent tissue.Conclusion:The expression of AR in HCC was correlated with the degree of tumor differentiation,and was significantly different in different degree of differentiation,which could be used as a new marker for predicting the prognosis of HCC.
