Study On Quality Standard For Weilening Tables And Its Drug Substance Hericium Mycelium With Solid Cultures

Weilening Tables are made from H.erinaceus with solid cultures.At present,Weilening Tables are the national protected variety of traditional Chinese medicine.And widely used in the clinical treatment of stomach pain,chronic atrophic gastritis,peptic ulcer,etc.Health Department standard(WS3-B-3457-98)of Weilening Tables and its drug substance H.erinaceus with solid cultures only has the properties and two chemical identification,which is lacking of feature.A systematic study was carried out in this thesis for the better quality standard for Weilening Tables and its drug substance H.erinaceus with solid cultures.Adenosine and amino acids(including leucine,valine,threonine,glycine,histidine)in Weilening Tables and H.erinaceus with solid cultures were used to identify by thin-layer chromatography(TLC).The TLC spots developed were clear.The samples were in the corresponding position with the reference herb and the reference substance respectively.According to the methods recorded in the Chinese Pharmacopoeia(2015 Edition),the water,water soluble extractive,ethanol soluble extraction,total ash,acid insoluble substance,heavy metals and harmful elements,aflatoxin in H.erinaceus with solid cultures were carried out.The content of 3 kinds of nucleosides(guanosine,uridine,adenosine)and 2 kinds of sterol(ergosterol,?-sitoster)in H.erinaceus with solid cultures were determined by HPLC.The data calculation was performed with Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(Version 2004A)to establish the HPLC fingerprint.The fingerprint of 10 batches of H.mycelium with solid cultures were carried out by similarity comparison,and 21 chromatographic peaks were extracted as the common peaks of fingerprint.The content of polysaccharide was determined by ultraviolet-visible spectrophotometry(UV-VIS).The content of adenosine and specific chromatogram in Weilening Tables were determined by HPLC.The specific chromatogram of 10 batches of Weilening tables were carried out,and 5 chromatographic peaks were extracted as the common peaks of specific chromatogram.The quality standard established,which can be used for the quality standard of H.erinaceus with solid cultures,is simple,accurate,reproducible,and specific.
