Background: laryngeal squamous cell carcinoma(laryngeal squamous cell carcinoma,LSCC)is one of the most common malignant tumor of head and neck,serious harm people's health,incidence increased year by year.Angiogenesis is closely related to tumor growth and metastasis,for every link of angiogenesis anti-angiogenesis therapy for the treatment of malignant tumor has brought new hope.Platelet response protein 1(TSP thrombospondin-1,)is first discovered angiogenesis inhibiting factor,through the analysis of impact on the regulation of angiogenesis of tumor growth and metastasis.Reported in the literature as a TSP-1 expression is associated with a variety of the clinical pathological characteristics and prognosis of cancer,and prove the expression of TSP-1 negative correlation with tumor angiogenesis.At present,the TSP-1 expression in laryngeal cancer and its clinical significance did not see related literature at home and abroad.But,as a kind of multifunctional proteins,TSP-1 gene expression may also be controlled by many factors,it is a complex process of invasion and metastasis of tumor to participate in,may play multiple roles in tumor occurrence and progress of,so to explore a new evaluation prognostic factors and relations with laryngeal squamous cell carcinoma of the TSP-1 is of great significance to the treatment of laryngeal squamous cell carcinoma.Objective:The aim of this study was to investigate the expression of thrombospondin-1(TSP-1)in laryngeal squamous cell carcinoma(LSCC)and to correlate the expression level with the biological behavior and angiogenesis of LSCC.Methods:Sixty-four patients(49 men and 15 women)with LSCC were enrolled in 2004–2006 and followed-up for 5 years.TSP-1expression in tumor tissues of these patients was detected immunohistochemically.The relationships between TSP-1 expression,prognosis,and clinicopathological features of the LSCC cases were analyzed.Further,CD105 immunohistochemical staining was performed in the same microarray,and was used to calculate the microvessel density(MVD)in LSCC tumor tissues.Finally,the correlation between MVD and TSP-1 expression was investigated.Results : 1 ? TSP-1 expression level in different organization.in normal tissues respectively is 96.55%,accounting for 60.94% of primary cancer,accounting for 45.00% of metastatic carcinoma and normal tissue expression rate is high.2?TSP-1 protein expression of quantitative normal tissue was 0.066 ± 0.022,respectively,0.038 ± 0.033 primary lung cancer,metastatic carcinoma of 0.017 ± 0.018,the expression of TSP in the normal tissue-1 quantity is obviously higher than that of primary lung cancer and metastatic carcinoma.Positive expression of TSP-1 on survival or express high levels of low expression is significantly higher than TSP-1 or low levels of patients.3?the expression of TSP-1 with laryngeal cancer were significantly correlated with the differentiation degree and the presence of lymph node metastasis and the expression of TSP-1 quantity well-differentiated group was obviously higher than in differentiated and poorly differentiated groups,the difference was very significant meaning(P < 0.001);The expression of TSP with lymph node metastasis group-1 quantity was lower than that in group without lymph node metastasis,significant differences(P < 0.05);The relationship between the expression of TSP-1 and tumor primary site,scope of infiltration and the patient's age,gender and other factors were no significant(P > 0.05).4?TSP-1 positive expression of 39 cases of MVD value 6.974 ± 6.974,25 cases of negative expression of TSP-1 average MVD was 11.420 ± 3.236,TSP-1 positive expression of MVD was significantly lower than the negative expression of group.Conclusions:Low expression of TSP-1 in LSCC was significantly associated with lymphatic metastasis and poorer differentiation of this disease.Further,low expression of TSP-1 in LSCC significantly correlated with increased angiogenesis in tumor tissues.TSP-1 expression deficiency may have contributed to the poor prognosis of some patients with LSCC.
