BackgroundAlzheimer's disease(AD)is one of the most common neurodegenerative diseases.its etiology is not yet fully illustrated for the pathogenesis is very complex.The incidence of AD increases with age,and women are more susceptible than men.Researches of Traditional Chinese Medicine are becoming research focus because of the therapeutic mechanism of their multiple targets in the mechanisms.Traditionally,Danggui Shaoyao San(DSS)is mainly used for gynecological diseases,but is widely used in many kinds of diseases in clinic at present,and there are a lot of clinical and experimental researches about treatment of nervous systemic diseases.In this study,we explored the potential targets of AD using DSS,which was based on the efficacy and modern pharmacology research.ObjectiveClinical and experimental investigations have demonstrated that estrogen decline leads to a high risk of the incidence of alzheimer's disease(AD),appropriately estrogen supplement therapies could attenuate disease-associated cognitive deficits.Previous studies have proved that Danggui Shaoyao san(DSS)have estrogen-like and neuroprotective effect.In this study,we explore the effect of DSS on metabolic disorder,cognitive impairment and hippocampal inflammation in ovariectomized(OVX)mice,which is commonly used to mimic the estrogen deficiency lead to cognitive deficits after menopause.MethodsOVX mice were administered with DSS for 12 weeks.During the treatment,the mice were weighed once a week.Memory function and anxiety-behavior were examined by Morris water maze,Y-maze and open field test.Finally,the plasma lipid levels were measured,and the number of microglia and astrocytes,protein expression of inflammatory mediator and two kinds of estrogen receptor in hippocampus were examined.ResultsWe found that DSS could reduce the increased body weight and relief the metabolism disorders induced by OVX.In addition,DSS could ameliorate the memory deficit and anxiety-behavior of OVX mice,and inhibit the glial activation in hippocampus,down-regulate the level of NF-?B,NLRP3,ASC,caspasel and IL-1? in hippocampus.This may be associated with ERs regulated.ConclusionOur data suggest that DSS significantly modulates estrogen deficiency-induced metabolic disorder,cognitive impairment,and its estrogen-like action can mitigate the hippocampal inflammation in OVX mice,which may by regulating the NLRP3 inflammasome pathway via ERs.Thus,DSS is a promising drug for the treatment of metabolic disorder and neurological symptoms in women around or after menopause.
