Purpose1.Study Asthma effect changes before and after the intervention of SAD and its singleherb compatibility.2.Study changes of plasma components before and after the intervention of SAD and its singleherb compatibility.3.Study the internal relationship between its efects and component contents before and after the compatibility of SAD and its singleherb;comment on the meaning of SAD compalibility.Method1 According to 'Ju Fang',the dose of SAD is ephedra:almond:licorice=1:1:1.Through the experiment with water extraction,extract the herb broth of SAD,ephedra-almond-licorice miscible liquids,single ephedra Decoction,single almond Decoction,single licorice Decoction for spare experiments.Research on components of plasma and herbs.Choose UPLC-MS to detect the components of plasma and herbs.Choose ephetonin,L-N-Methylephedrine Hydro Chloride,halofed,formononetin,amygdaloside,prunasin,glycyrrhetinic acid and liquiritin to perform related experiments and get standard solution.Explore the liquid phase condition and determine the standard curves.Give the enema to rats with miscible liquids group,single ephedra group,single almond group and single licorice group for three consecutive days,collect and purify the plasma.By the same method prepare the same sample solution and determine the content,get relevant atalas,content results,analyze the components by the internal standard method and the data.2 Research on effect evaluation.Choose miscible liquids group,single ephedra group,single almond group,single licorice group to perform experiments.Copy OVA mouse model of asthma,collect processing data of airway hyperresponsiveness,observe and evaluate the lung biopsy of mouse,discuss the effects different groups have on mice's asthma and give objective comments.ResultEffect of intervention on asthma s uggests:compare to the model group,SAD and ephedra-almond-licorice miscible liquids both can decrease the level of EOS in the blood(density<0.05 or<0.01),improve lung resistance(penh)and lung pathological model of asthmatic mice.Single ephedra Decoction,single almond Decoction and single licorice Decoction have no apparent improvements on the number of eosinophils in blood,lung resistance and pulmonary pathology.Content of enter-blood components suggests:It's better to separate ephetonin,L-N-Methylephedrine Hydro Chloride,halofed,formononetin,amygdaloside,prunasin,glycynetinic acid and liquiritin.And components have specificities and are easy to identify.Ephetonin,L-N-Methylephedrine Hydro Chloride and halofed have highest blood-dr ug content in SAD,same blood-dr ug content in miscible liquids and single ephedra goup;glycyrrhetinic acid,liquiritin and formononetin have highest content in SAD,miscible liquid group is slightly higher than single licorice group.Conclusion1.Prescription analysis of SAD believes that ephedra,diaphoresis,disperses lung qi and relieves asthma;bitter almond descendes lung qi and phlegm laxative;licorice slightly reinforces the spleen and remove phlegm,heat and harmonic resistance.The three are of special significance in the compatibility of drugs,lifting,process:ing and so on.2.Experiment proves:SAD boiled together can reach the maximum benefit along with highest blood-drug content;ephedra-almond-licorice miscible liquids have certain effects on intervention of asthma along with comparatively lower blood-drug content More active ingredients and it functions more strongly.3.Licorice compatibility may have biological regulation and it's helpful to improve the bioavailability of drugs,delay the metabolic time between themselves and drugs,and enhance the efficacy.4.In the process of SAD's intervening asthma,ephedra,almond and licorice are in separable and overall have efficient results for intervention of asthma.Furthermore,contents in the blood reach to highest level after being boiled together.It is the main reason for antitussive and antiasthmatic,detoxification anti-in flammatory effect of strong reaction,reflecting the correctness of the compatibility of the Traditional Chinese Medicine theory.
