Diabetes has now become the third largest disease following cardiovascular and tumor.There is a lot of treatment for diabetes in the market.But there is a certain adverse reaction.For exmple,insulin will causes low blood sugar in patients.Western medicine treatment of diabetes is targeted,but there are certain liver and kidney toxicity,limiting its use in the treatment of diabetes.The treatment effect of traditional Chinese medicine on diabetes has a long history.It has little side effects and can be taken for a long time.Therefore,in recent years,the hypoglycemic activity of traditional Chinese medicine has received extensive attention and made good progress.Physalis is rich in the Changbai Mountains,and the main distribution is in the Inner Mongolia area and the three northeastern provinces,especially Jilin province.This paper uses Physalis as experimental materials,studying the extraction,purification and tablets process of Physalis alkekengi calyx saponins,and hypoglycemic activity.In this paper,the purification process of Kam lantern is optimized.Through the single factor test,the extraction and purification process were optimized by orthogonal test on the basis of obtaining the best single factor extraction condition.The optimum extraction conditions were alcohol concentration 70%,extraction temperature 70 ℃,solid-liquid ratio 1: 6,extraction time 0.5 h,extraction times 4 times,and its optimized extraction rate reached about 4.8%.The optimum purification process was water elution volume 4BV,purification solvent concentration 70%,loading concentration 60 mg / ml,optimized saponin yield reached about 70%.Secondly,the optimal process for the preparation was as follows: dextrin: starch: lanterns calyx saponins: ethanol = 7:7:1:3.Mixture,wetting agent selected 50% Ethanol,and lubricant magnesium stearate 0.5% is more appropriate.The quality of the tablets,such as the appearance,the difference of tablets,the time of disintegration,the dissolution rate,the heavy metal and the residual of organic matter,were in accordance with the regulations.On the basis of the above research results,this paper evaluated the hypoglycemic activity of pills in lanterns(TSP)tablets.The differences between the TSP low,medium and high dose group and the Xiaoke pill group were not significantly different.After 14 days of tonic injection,the TSP low,middle and high dose group were compared with Xiaoke pill group,and the difference was significant(P <0.05).Blood glucose,sugar tolerance were significantly reduced.14 days of the test results show that food intake,drinking water,excretion,weight have been a certain degree of improvement.The above results provide a theoretical basis for the further development of hypoglycemic used as functional foods or drugs.
