Objective: We profiled the expression of miRNAs in serum related to OMLDT and evaluated the potential of abnormal expressed miRNAs as precise diagnostic markers.Methods: Serum samples from 5 patients with OMLDT and 5 trichloroethylene(TCE)contacts were collected.The total RNA was extracted then labeled by the Hy3 fluorescent.microRNAs were hybridized with miRCURY LNATM chips and images were acquired using the GenePix 4000 B scanner.SYBR-PCR method was used to verify the results of miRNA microarray include 34 patients with OMLDT and 34trichloroethylene(TCE)contacts.MGB-PCR method was used to verify the results of SYBR-PCR.Associations between relative expression of miRNAs and liver function indexes were analyzed using the Spearman correlation test.The diagnostic potential of the miRNAs were analyzed using SVM algorithm.A database of differentially expressed miRNAs were Constructed.Data were treated by normalization.Randomly divided 60% of the sample(40 cases)into a training set,the rest for the testing set.The best parameters of model was founded by tripled cross validation.The diagnostic model of occurrence of medicamentosa-like dermatitis induced by trichloroethylene was constructed by Support Vector Machine algorithm(SVM).Results: A total of 69 microRNAs were differentially expressed between OMLDT and TCE contacts,in the microRNAs,39 were up-regulated and 30 were down-regulated.hsa-miR-193b-3p ? hsa-miR-339-5p ? hsa-miR-10a-5p ? hsa-miR-34a-5p ?hsa-miR-21-5p were verified increased in OMLDT using SYBR-PCR.hsa-miR-339-5p and hsa-miR-21-5p were verified increased in OMLDT using MGB-PCR.Liver function indexes including TP?ALB?DBIL?IBILI?ALT?AST?GGT?ALP were statistical differences between patients with OMLDT and control.Correlation analysis showed that hsa-miR-21-5p was negatively associated with TP,ALB and GLB however positively associated with TBIL,DBIL,ALT,AST and GGT.hsa-miR-339-5p was negatively associated with TP,ALB and GLB.The sensitivity,specificity and accuracy of SVM diagnostic model were 100%,75% and 85.71%% respectively.CONCLUSION: The findings of this study show that the expression of microRNA in serum ofpatients with OMLDT has significantly altered,and miR-21-5p and miR-339-5p could serve as candidate biomarkers for the identification of OMLDT.The diagnostic model has a good goodness of fit and prediction accuracy of OMLDT occurrence.
