Objective:artificial surgery cause jaundice mice as a model to moderate intensity exercise and HWTX-11 tail vein injection of intervention.Observation HWTX-11 Joint intervention of moderate-intensity aerobic exercise for miceobstructive jaundice explore obstructive jaundice secondary liver injury and the possible molecular mechanisms of action.Methods:50 KM mice were randomly divided into 5 group:Heath mice,control group,HWTX-XI group(0.3(?g/g),the aerobic exercise group and the aerobic exercise and drug in combination group(the combination group).In the common bile duct to the abdominal wall hanging 48 hours to build miceobstructive jaundice caused by severe liver injury model.Judging from the low-dose group of high school blood bioAKPmical indicators of optimal dose,in order to participate in the exercise and HWTX-11 in the combination therapy group.Application of hematoxylin-eosin(HE staining)of each group mice liver tissue injury situation microstructure morphology;Measurement Alb,R-GT,AKP bioAKPmical methods,TBIL content,immunohistoAKPmistry to detect liver tissue TNF-?,IL-6,NF-?B protein expression;RT-PCR technology to detect TNF-?,IL-6,NF-?B expression.Results:?HE staining:Healthy mice hepatic cord arranged in neat rows;control group mice hepatocytes lot of fibrosis:showing a large sheet of liver cell degeneration,necrosis,and even a nuclear substance isolated vacuoles;HWTX-11 group,the simple exercise group visible liver cell a few points,necrosis,lobular structural changes,hepatic cord neatly arranged owe;HWTX-11+exercise group had no significant tissue necrosis,lobular structure normal hepatic cord arranged in neat.?serological results:HWTX-11+exercise group TBIL,AKP,R-GT levels than control group significantly decreased(P<0.01),HWTX-11 group,exercise group TBIL,AKP,R-GT levels were also significantly decreased(P<0.05).?immunohistoAKPmistry show TNF-a,IL-6,NF-?B positive minimum expression;liver tissue TNF-a,IL-6,NF-?B expression lower than the model control group.?RT-PCR results showed that:with the control group comparison,HWTX-11+exercise group mice serum TNF-a,reduce NF-?B,IL-6 expression(P<0.01).Conclusion:Moderate intensity aerobic exercise intervention of the United HWTX-11 tail vein injection of miceobstructive jaundice,and can enhance the obstructive jaundice caused by severe liver injury mice's immune function,thereby significantly reduce the degree of obstructive jaundice induced mice liver injury.
