Objective:In this study,we collected bone marrow cell samples from clinical acute myeloid leukemia and cultured human acute myeloid leukemia cell lines which had been to observe the expression of transient receptor potential vanilloid receptor five and transient receptor potential vanilloid receptor six channel in acute myeloid leukemia(AML),as well as the correlation between TRPV5 and TRPV6 expression levels and clinical features of AML patients.Methods:To extract the protein and total RNA for experimental group in AML patients with bone marrow mononuclear cells and AML cell lines,for the control group mobilized after bone marrow mononuclear cells.the mRNA expression of TRPV5 and TRPV6 in AML was detected by quantitative real-time polymerase chain reaction(qRT-PCR),and western blot was used to detect the protein expression of TRPV5 and TRPV6 in AML.We analyzed the relationship between TRPV5 and TRPV6 expression and clinical features of the patients,including the percentage of bone marrow blast cells,peripheral white blood cell count,hemoglobin,platelet count,sex,age.Results:The expression of TRPV5 and TRPV6 were significantly higher in initial treatment group,relapse group,cell lines group than in control group(P<0.05).The expression of TRPV5 and TRPV6 in complete remission(CR)group showed no significant difference from in control group(P>0.05).The expression of TRPV5 and TRPV6 in initial treatment group and relapse group AML had a positive correlation with the cell percentage of original naive bone marrow cells(P<0.05),but no obvious correlation with the patients' peripheral white blood cell count,hemoglobin,platelet count,sex,age.(P>0.05).Conclusions:(1)The mRNA and protein levels of TRPV5 and TRPV6 were up-regulated in initial AML,relapsed AML and AML cell lines.(2)There was no significant difference in TRPV5 and TRPV6 expression levels in complete remission of AML.(3)In initial and relapsed AML,the expression levels of TRPV5 and TRPV6 channels were positively correlated with the percentage of bone marrow cells in bone marrow.(4)In initial and relapsed AML,the expression levels of TRPV5 and TRPV6 were not associated with peripheral white blood cell count,hemoglobin,platelet count,age,sex,and bone marrow chromosome karyotype.
