ObjectiveThis study were carried on the hyperlipidemia rats which caused by high-fat diet,in order to analysis the effect of ethanol extracts from Spatholobus Suberectus on lipid and antioxidant enzymes,which exist in blood and liver.Pathological study was also done to observe the hepatoprotective effect and to invest its mechanisms and active ingredients.Methods1.Purification and screening the inhibition part of procyanidins from Spatholobi caulis by pancreatic lipase1.1 Determination of procyanidins and optimization of the extraction process Optimize the chromogenic condition by concentrated sulfuric acid-Vanillin.1.2 Study of extraction and purification of procyanidins from Spatholobi caulisOptimize the extraction technology of Procyanidins from spatholobus caulis by using single-factor test and determine the content in different periods.Purify the crude extract of Procyanidins from spatholobus caulis by using the macroporous resin.1.3 Study of the Proanthocyanidins' inhibition on pancreatic lipaseEstablish the method of Proanthocyanidins'inhibition on pancreatic lipase,then determine the purity of different elution and observe the inhibition of pancreatic lipase activity.1.4 The study of the effect of purification of procyanidins from Spatholobi caulis on macrophage comes from mice induce by LPS to inflammationEstablish the inflammation model,and observe the effect of Alcohol Extracts from Spatholobus suberectus Dun,purification of procyanidins from Spatholobi caulis and catechin.2 The effect of Spatholobi caulis on lipid metabolism and antioxidation level in ratThirty-six SD rats were randomly divided into control group,model group,positive control group,low-dose group,middle-dose group,high-dose group(2?4?8g·kg-1·d-1)of Spatholobi caulis.After treatment for 12 continous weeks,the rat were killed with anesthesia,collected the liver,monitored the lipid level,enzyme activity and pathological changed in liver.Resu!t1.Purification and screening the inhibition part of procyanidins from Spatholobi caulis by pancreatic lipaseBest color condition:20g/L vanillin,30%sulfuric acid,reacted 15min at room temperature stay away from light.Best extraction is:80%alcohol reflux extraction in solid-liquid ratio of 1:15 to 90 min.May has the highest content of proanthocyanidins in twelve months.X-5 macroporous resin was selected by static adsorptionand resolution to purify procyanidins from Spatholobi caulis.The flow rate was 2BV/h and the sampling amount was 8BV.Washed out by different volume fraction of ethanol have different purity of proanthocyanidins,inhibition of pancreatic lipase activity was also different.Purified washed out by 50 percent of ethanol was the most pure one,94.67T,and has the highest inhibitional rate of all purified.Purity has a positive correlation with inhibition.Alcohol Extracts from Spatholobus suberectus Dun,purification of procyanidins from Spatholobi caulis and catechin have anti-inflammatory effect on macrophage comes from mice induce by LPS to inflammation,and the first one has a better effect than the other two.2.The effect of Spatholobi caulis on lipid metabolism and antioxidation level in ratIn this experiment,Spatholobi caulis significantly reduced serum TC,TG,LDL-C,AI,and R-CHD,significantly increased HDL-C,HDL/TC,compared with the model group.At the same time,it significantly increased serum CAT enzymatic activity,but it doesn' t have significant effect on NO,AKP,and SOD.In the aspect of liver,Spatholobi caulis significantily reduced liver TC,TG,and increase GSH-PX enzymatic activity,but doesn t have effect on CAT.High dose group of spatholobus caulis has a significant hepatoprotective effect.Pathological section show that alcohol extracts from Spatholobus suberectus Dun has hepatoprotective effect too.ConclusionSpatholobi caulis crude extracts and purified on inhibiting pancreatic lipase inhibition was positively correlated with purity.Spatholobi caulis crude extracts has better anti-inflammation effect in vitro than purified Ethanol extracts from Spatholobi caulis significantly reduced lipid levels and increasing peroxidase activity,with good prevention of hyperlipemia and fatty liver.
