Over the past 30 years,as a result of the changes of diet and lifestyle,the incidence of gouty arthritis increased drastically in China.However,no effective and affordable drug for the treatment of gouty arthritis has been developed.As a traditional Chinese medicine,Spatholobus suberectus Dunn has been used for the treatment of rheumatoid arthritis with other traditional Chinese medicine.In the first chapter of this thesis,progresses on gouty arthritis,the chemical constituents and the pharmacological activities of Spatholobus suberectus Dunn were reviewed.In the second chapter,a gouty arthritis model in vivo was established by injecting monosodiumurate(MSU)crystal into the toes of mice,which was then employed to evaluate the biological effects of the aqueous extracts of Spatholobus suberectus Dunn.The results showedthat,as compared with the control group,the aqueous extracts of Spatholobus suberectus Dunn could reduce the swelling of mice toes with the high-dose group significantly reducing the swelling effect(P?0.01 as compared with the control group),which indicated that the aqueous extracts of Spatholobus suberectus Dunn might contained active constituents for the treatment of gouty arthritis.In the third chapter,chemical constituents in the aqueous extracts of Spatholobus suberectus Dunn were investigated.As a result,ten compounds were isolated and identified by a comprehensive application of column chromatography(silica gel,ODS C18,Tosh HW-40,etc.)and spectroscopic analyses(NMR and MS),including a new compound 3 and nine known compounds 1-2,4-10.Compounds 1-6 were flavonoids and compounds1,9 and 10 were isolated from the genus Spatholobus for the first time.In the fourth chapter,the anti-inflammatory activity of compounds 1,2,8,4and 5 in vitro were evaluated.As a result,the five compounds exhibited potent anti-inflammatory activity.The results of the study suggested that the aqueous extracts of Spatholobus suberectus Dunn warrant further study.
