Thais clavigera Kuster is a species of the genus Thais of the Muricdae family,which is distributed in our country,north and south coastal,Japan,Korea and Viet Nam.Modern pharmacological studies have shown that the extract of Thais clavigera Kuster has good activities such as antibacterial,immunomodulation and anti-tumor.In recent years,many studies focused on the development and utilization of ocean bio-resources was be reported,there are many chemical constituents from Thais clavigera Kuster have been reported,including triterpenoids,coumarins,flavonoids and steoids.The present study is undertaken further on the chemical constituents of Thais clavigera Kuster,in order to find more compounds with medical values and better comprehensive development and utilization.This article first studied the organic solvents part of Thais clavigera Kuster.The meat of Thais clavigera Kuster was extracted three times with methanol.The extract was partitioned with petroleum ether,ethyl acetate and n-BuOH.18 chemical compounds were isolated by means of silica gel column chromatography,recrystallization and preparative thin layer chromatography.Based on the physical and chemical properties of the compou-nds,nuclear magnetic resonance spectroscopy(1H-NMR,13C-NMR)and mass spectrometrydata,the chemical structures of 11compounds were identified after compared with the literatures.The compounds isolated from Thais clavigera Kuster were elucidated as:5-hydroxyl-3,4-dimethy-5-pentyl-2?5H?-furanone,1H-indole-2,3-dione,6-bromo-,1H-Indole-3-methanol-5-hydroxy-2-methyl,Octadecanoic acid,2H-indol-2-one,6-bromo-1,3-dihydro-3-hydroxy-3-?2-oxopropyl?-,?S?-3-dodecyloxypropane-1,2-diol,Cholesterol,Benzamide,2-amino-N-?4-amino-4-oxobutyl?-4-bromo-.This article also studied the Water-soluble part of Thais clavigera Kuster.Crude polysacharides were isolated from Thais clavigera Kuster by a method so called“hot water extraction followed by ethanol precipitation”,Single factor test showed that the optimal conditions for extraction of polysacharides from Thais clavigera Kuster were as follows:temperature-70?,solid-liquid ratio-1:30,extraction running times-2 h.The polysaccharide content was determined by phenol-sulfuriic acid method.Finally,the content?percentage?of total sugar was determined to be?8.35±0.42?mg,the ratio of extraction was?0.17±0.01?%.By comparing enzymolysis to remove protein showed that,it removed protein at?60.26±1.12?%.After ethanol precipitation drying,its content of total sugar was?48.17±0.17?%,protein content was?31.85±0.42?%.The polysaccharides of Thais clavigera Kuster possessed good antioxidant activity in vitro as evaluated by scavenging assays involving hydroxyl radical?·OH?and1,1-diphenyl-2-picrylhydrazyl?DPPH·?,IC500 respectively 6.31 mg/mLand 2.25 mg/mL.
