Background and PurposeWith the effect of attacking stagnation,purging intense heat and detoxification,rhubarb and purgative preparations containing rhubarb have been widely used in clinical practice.Sennoside,as the main component of rhubarb,its primary mechanism of diarrhea was that sennoside had been degraded by gut microbiota into corresponding Flavonids in the body,which stimulated the intestinal wall and promoted intestinal peristalsis,then led to diarrhea.In previous laboratory study,it occasionally proved that the use of sennoside A could increase the Akkermansia muciniphilaon of mouse's bowel flora.Through excavating literature,Akkermansia muciniphila has been found to play important role to lower blood glucose.These results have aroused our interests greatly.Therefore,this paper aims to investigate the hypoglycemic effect of sennoside A and the relativity between its hypoglycemic mechanism and gut microbiota,so as to provide experimental and theoretical guidance for the study of gut microbiota and the mechanism of Chinese medicine treatment of disease.Research Contents and ResultsThis study adopted the second generation sequencing technique to detect the gut microbiota of db/db diabetic mice under normal and hyperglycemic phenotype,and research the changes of gut microbiota before and after the diabetic phenotype.The results showed that gut microbiota changed dramatically after db/db diabetic mice under hyperglycemic phenotype.It showed that the number of Firmicutes bacteria increased significantly,and the proportion of Firmicutes and Bacteroides also rised.Animals acted as bulimia and excessive obesity,while some bacteria with the effect of hypoglycemic(especially Akkermansia muciniphila)significantly reduced.Due to the disorder of gut microbiota structure in diabetic mice,we investigated the hypoglycemic effect and the mechanism of hypoglycemia of sennoside A,which had effect on regulating gut microbiota.The results showed that sennoside A could significantly change the gut microbiota structure,reduce the Firmicutes flora and decrease the proportion of Firmicutes and Bacteroides,then it come into being obvious hypoglycemic effects.In order to verify the possible mechanism of sennoside A hypoglycemic mechanism,fecal transplantation method had been adopted to study the hypoglycemic effect of gut microbiota.It was found that the gut microbiota of each administered group could change the gut microbiota of db/db mice,decrease the proportion of Firmicutes to Bacteroides,then have a significant hypoglycemic effect.Conclusion and DiscussionAccording to the above research contents and results,it's not hard to find that diabetes in db/db mice has a significant association with gut microbiota.Sennoside A could change the gut microbiota of db/db mice and come into being the effect of lowering blood glucose,which showed that the lowering blood glucose mechanism of sennoside A may have relationship to the regulation of gut microbiota.The method of fecal transplantation was further to verify the effect of lowering blood glucose of sennoside A depended on the gut microbiota.This study fully demonstrates that the effect of lowering blood glucose of sennoside A may through regulating the mechanism of gut microbiota.This study revealed that the mechanism of hypoglycemic effect of rhubarb perhaps was related to the regulation of gut microbiota,which provides experimental and theoretical basis for exploring the way of traditional Chinese medicine to treat diseases.
