| Currently,SM formula has been clinically used for treating cardiovascular and cerebrovascular diseases.Further,lots of researches have demonstrated that SM has a significant activity on scavenging free radical and battling against lipid peroxidation.Meanwhile,the loss of redox homeostasis in the body is closely linked to many kinds of human deseases,and over anti-oxidant treatment is believed to be harmful to the body.However,SM formula as an anti-oxidant agent has not been investigated on the respect of maintaining the redox homeostasis of the body.In present work,oxidative damage mouse model induced by the D-galactose was used to evaluate the antioxidant activity of SM formual and reduced SM formula.The content of MDA,the activity of SOD and GSH-Px has been selected as antioxidant indice to be detected in the tissues of mice.The result suggested that SM significantly promoted anti-oxidant capacity of the body,even excessed that in the normal mice,thus leading the body to be in a state of excessive reduction.While reduced SM significantly increased antioxidant activity in model mice to the level of normal mice.Taken together,reduced SM has been selected as a potential antioxidant health product for further development.Secondly,the acceptable forms of reduced SM crude powder and reduced SM ethanol extract have been revealed by single factor and orthogonal experiment.Reduced SM crude powder will finally be prepared in form of granules with 8% of potato starch as the adhesive and 3% of low substituted hydroxypropyl cellulose as the disintegrating agent.Accordingly,three batches of reduced SM granules were prepared,and have completely satisfied to the requirements of the Chinese Pharmacopoeia.Reduced SM extract powder will be made into capsules with the reduced SM extract powder,potato starch and low substituted hydroxypropyl cellulose at a dosage ratio of 10:1:0.2.Three batches of capsules were also accordingly prepared,and have met the requirements of the Chinese Pharmacopoeia.Finally,marker substances of total saponins and total flavonoids in reduced SM extract have been analyzed by spectrophotometry.In addition,the ginsenosides and Ophiopogon flavanone B have detected by HPLC.In present work,total flavonoids and total saponins were selected to be as marker substance of Radix Ginseng and Radix Ophiopogonis,respectively,and their contents will be determined by spectrophotometric method for the quality control of redued SM.In conclusion,reduced SM significantly increased anti-oxidation activity in oxidative damage mice induced by the D-galactose,reduced SM crude powder and ethanol extract have been prepared in dosage form of granules and capsules,respectively.Total flavonoids and total saponins were selected to be as marker substance of Radix Ginseng and Radix Ophiopogonis for redued SM quality control.The results will provide data and theoretical base for the development of reduced SM to be an anti-oxidant health product. |
