| At present,some studies have shown that Capparis spinosa L polysaccharides can induce tumor cell apoptosis through mitochondrial pathway to inhibit tumor growth.Selenium polysaccharide has better anti-tumor activity,some researchs have proved that selenium polysaccharide extracted from mushrooms,fungus and other plants have a better anti-tumor activity.Selenium polysaccharide both have the anti-tumor activity of Selenium and Polysaccharide,while reducing the toxicity inorganic selenium.Therefore,this study as a starting point in-depth research,research the Inhibition of Capparis spinosa L selenium polysaccharide(Se-CSPS)on human gastric cancer cell SGC-7901.So this article explore the correlation of some sides such as the mitochondrial membrane potential,concentration of RoS and Ca2+,between Se-CSPS and apoptosis of SGC-7901.Then clarify the Se-CSPS induced tumor cell apoptosis molecular mechanism,to further to Se-CSPS based research and development of anti trunor new drugs for certain experimental basis.At first this issue examines the inhibition of Se-CSPS for SGC-7901 cell growth by Inverted microscope,Fluorescence microscope and MTT assay,experimental results show that with the concentrations of Se-CSPS increasing,SGC-7901 human gastric cancer cell growth inhibition rates will also rise,both with dose-related;Observed under an inverted microscope,cells were suspended after the administration,and with the higher concentration of Se-CSPS,the longer duration of action,the phenomenon is more obvious;The results observed under fluorescent microscope show that the number of apoptotic bodies increased as the concentrations of Se-CPSP elevated.In Further research,SGC-7901 cell apoptosis was observed in case of different concentrations of Se-CPSP by flow cytometry PI staining method,the results show that,with increasing concentrations of Se-CSPS apoptosis also increased,the results show that with the increase of the concentration of Se-CSPS apoptosis rate of human gastric cancer SGC-7901 cells also increased,showing a positive correlation between them.Laser scanning confocal microscopy to detect cell reactive oxygen species and mitochondrial membrane potential calcium levels found thate with the concentrations of Se-CPSP increasing,the concentration of reactive oxygen species and calcium ion elevated,the level of mitochondrial membrane potential reduced.To sum up,Se-CSPS can induce apoptosis in human gastric cancer SGC-7901 cells through mitochondrial pathway. |
