Purpose This study uses Erzhi Recipe after establishing the intervention model of rats with degenerative cervical disc for cytology and genetics experiments.Surgry was applied to get cervical disc and nucleus pulpous tissue in rats and immunohistochemical staining techniques to observe Erzhi Recipe on the expression of MMP-11 in degenerative cervical disc and apoptosis in the nucleus.It aims to investigate the mechanism of expression of MMP-11 in cervical nucleus pulposus intervented under by Erzhi Recipe.Method A systematic review of the understanding of etiology and pathogenesis of cervical spondylosis in Chinese Medicine was undertaken firstly,while contrasting the research progress and understanding of cervical spondylitis of modem medicine.The experimental design was firstly randomized and then parallel-grouped.No deaths of mice was found in the acute toxicity tests,of Erzhi Recipe was not obtained,40 SPF ICR mice,weighing 18-20g,were randomly divided into A and B two groups with 20 mice in each.The recipe was concentrated to the maximum concentration and repeatedly administered to the mice of two groups of 3 times/day for Group A and 4 times/day for Group B.The mice was fed.4h after the end of the last administration,continuous observation of the animal changes was carried onfor seven days.The mice were weighted after the 7th day and then killed by cervical dislocation,dissected to observe the changes in the appearance of the internal organs of mice.Then experiment of the effect on expression of MMP-11 gene in cervical degenerative disc after using Erzhi Recipe was undertaken:80 SD rats,weighting 180-200g,were divided into A,B,C,D groups with 20 in each.All the mice were taken cervical disc puncture,fed in groups after modeling succeed.A group(control group):modeling underwent saline;Group B(experimental group):after modeling fed the same dose of Erzhi Recipe.Group C(WM group)after modeling fed the same dose of diclofenac;D group(Chinese medicine group)after modeling fed the same dose of neck pain particles.30d after administration to take appropriate puncture disc tissue,making detection of MMP-11 and apoptosis observed nucleus pulposus.Result After one week of continuous intragastric administration,there were no deaths in acute toxicity test in two groups of mice.Within 1-2 days after the mice were intragastric administration the diet appears to reduce,lower activity levels,deereeased flexibility,then gradually returned to normal,activity sensitive,shiny fur,eating good observation found no systemic toxicity.Weighting all the mice after the last intragastric administration,weight did not change significantly.After the mice were sacrificed,then dissected and observed mouse kidney,stomach,spleen,lung,heart,liver,intestines and other organs,showed no obvious physiological and pathological changes.In the rat experiments,obtained through detection of MMP-11 gene expression level in the experimental group is lower than the control group(P<0.05),were significant.Conclusion Erzhi Recipe concentrated in the largest concentration is difficult to measure the acute toxicity median lethal dose,indicating that Erzhi Recipe is safe and nontoxic.Also lay a good foundation for the future to further evaluate the clinical safety.Under Erzhi Recipe can significantly reduce levels of MMP-11 gene expression,inhibition of apoptosis,and restoring the normal function of the intervertebral disc.The experiment is a more in-depth study of the mechanism of disc degeneration provides an experimental basis.
