[Object]A rapid,sensitive UPLC-TOF-MS/MS assay has been developed to study the fragmentation regularity from electrospray ionization mass spectra of picroside ? with positive and negative mode by fragment ions and references;According to the speculate of metabolites speculate the possible metabolism pathway of picroside ? from bile,urine,feces and intestinal flora.[Method]Picroside ? and metabolites were isolated on Inertsil ACQUITY UPLC(?)BEH C18 columns with a mobile phase of acetonitrile-0.1%formic acid.Electrospray ionization(ESI)source was used at both positive and negative models.Inferred the Picroside? fracture and fragmentation pathway manner with Massfragment software and the data by electrospray ionization positive and negative models;Picroside ? was incubated with rat intestinal flora in vitro under lack of oxygen;Identification and structural elucidation of the metabolites were performed by comparing the full scan MS and MS/MS spectra of analyzes with those of the parent drug and blank sample;After taking orally solution of picroside ?300mg/kg,and then bile from different time,(0-24h)urine and(0-24h)feces were pretreated and analyzed by UPLC-TOF-MS/MS,and to detected the mass information of metabolites,speculation and confirm structure of picroside ?.According to the speculate of metabolites speculation the possible metabolism pathway of picroside ? from bile,urine and feces;[Results]Picroside ? fracture mode and fragmentation pathway was inferred with UPLC-TOF-MS/MS mass spectrometry data and massfragment by electrospray ionization positive and negative models.Intestinal flora incubated experimental:We found 8(M1-M8)metabolites of Picroside ? in rat intestinal flora,and infer the possible metabolic pathway of Picroside ?.We detected metabolites of Picroside ? for biological samples in vivo accorded to UPLC-ESI-TOF-MS/MS analysis.We detected 2(M9 and M8)metabolites and parent drug in rat bile;and detected 5(M11-M15)metabolites in rat urine and feces,and no detected parent in rat feces,and infer the possible metabolic pathway of Picroside ? in vivo.[Conclusion]We found metabolites.And infer the possible metabolic pathway of Picroside ?in vivo and vitro by structures of metabolites.Picroside ? occurred extensive metabolism in in vivo and vitro but metabolic irregularities.The method is rapid,sensitive,simple and suitable for metabolic analysis of picroside ?;It was helpful to study the transformation pathways in vivo.
