Objective:To explore SENP1's role in the differentiation of the glomerulus,and to observe expression characteristics of SENP1 during murine metanephron development.Methods:RT-PCR technique was used to measure the expression level of SENP1 mRNA for the embryonic mouse E13.5d?E14.5d?E16.5d?E18.5d and after birth mouse Pld?1w?2w?4w,western blot was used to assess the protein expression changes of SENP1 and HIF-2a in the developing murine kidney from 13.5-18.5 to post coitom and postnatal day P1d?1w?2w?4w,immunohistochemistry and immunofluorescence staining were used to examine the expression locations and observe the expression characteristics of SENP1 protein at different stages of emoryonic and adult mice kidney.Results:During the development of the murine kidney,there is no significant different between all the groups,the expression level of SENP1 mRNA remains stable(p>0.05);compared to E13.5d group,the protein expression level of SENP1 reduced from E14.5d group and kept at the very low level after new born(p<0.05),the protein expression level of HIF-2a also reduced from E14.5d group and kept at the very low level after new born(p<0.05);the expression locations was too weak to be observed in both immunohistochemistry and immunofluorescence staining.Conclusion:During the development of the murine kidney,SENP1 and HIF-2a both decreased from E13.5d to P4d.They had a time consistency.
