Experimental Study On The Astragalus Extract Promote HaCaT Cell Proliferation

Objective:To study the astragalus extract on human keratinocyte cell line(HaCaT cells)proliferation and its possible mechanism,to observe HaCaT cell proliferation and NRP-1 expression under the effect of Astragalus extraction,Exploring its possible mechanism in the healing process of diabetic foot.Methods:1.Through MTT method to observe the effect in different concentration of Astragalus Extract(0,0.1,0.5,1,5,10,20,50,100mg/ml)action on the proliferation of HaCaT cells;2.RT-PCR method was used to observe the effect of the expression of NRP-1 mRNA in HaCaT cells under the 5mg/ml of Astragalus Extract;Through western blot observe NRP-1 protein expression level on HaCaT cells.under the 5mg/ml of Astragalus Extract.Results:1 Astragalus extract dependence concentration promoting HaCaT cell proliferation the optimum concentration is 5mg/ml;2 Astragalus extract has the effect on promoting the proliferation of HaCaT cells,probably relate to with the Astragalus Extract promote the NRP-1 protein expression on HaCaT cells.2.The 5mg/ml of astragalus extract could promote NRP-1 mRNA expression levels and the expression level of NRP-1 protein HaCaT cell.Conclusion:Astragalus extract has the effect on promoting the proliferation of HaCaT cells,probably relate to with the Astragalus Extract promote the NRP-1 protein expression on HaCaT cells.
