Effects Of Effective Components Of Qing Guang An On Inoblast And Type ? Collagen Of Scarring Tissue Of Filtration Canal After Glaucoma Surgery

Object:To observe effects of four effective components of Qing Guang An on inoblast and type ? collagen of scarring tissue of filtration canal after glaucoma surgery,and discuss their effect and function mechanism on preventing its formation,and a preliminary judgment on the safety of effective components of Qing Guang An will be given.Methods:42 healthy adult New Zealand long ears white rabbit,which average weight are 1.8 to 2.2kg,were divided into 7 groups randomly with 6 rabbits in each group.These seven groups were used as following:group A was blank group;group B was model group;group C was a control group of MMC;group D,E,F,G was used effective component signed respectively as 1,2,3,4.Rabbits' eyes in group B,C,D,E,F,G had conventional trabeculectomy and iridodialysis;group C use with MMC;do nothing on group A.The next day after molding began to use medicine on each group.Each group of experimental animals' intraocular pressure was measured after surgery,and the formation of filtering bleb was observed,and the intertion status and eyes' general condition were checked.After using medicine for four weeks all animals for experiment were executed,the whole organization level of filter canal were taken from eyes that have gain surgery.After dyed by HE organization structure where the surgery had done and the number of inoblast were observed.Expression of type I collagen in Tenon bourse in filtering bleb area was checked with the immunohistochemical method.Result:After surgery C,E groups of intraocular pressure rose slowly.At the fourth week intraocular pressure remained minimum.The compare of these two groups of intraocular pressure and those of group A,B,D,F,G had significance on statistics.C,E groups remained effective filtering bleb.Filtering bleb in group B turned into no function or disappeared two weeks after surgery.Group D,F,G had part of filtering bleb existing four weeks after surgery.Comparing with the number of inoblast of each group,except group D and E,which result was P>0.05 and had no obvious significance on statistics,others were significant.Comparing with the the expression of type I collagen,except group C and E,B and G,F and G? result of others have obvious significance on statistics showing as P<0.05.According to the observation of common condition after surgery and specimen polarizing at the fourth week,the group with MMC had complication after surgery,other groups with effective components did not show any abnormal lesions in eye organization structure.Conclusion:(1)The scaring of filter canal after glaucoma surgery is main cause of the filtration surgery's failure assuming abnormal backup intraocular pressure.(2)In doing conventional trabeculectomy and iridodialysis,a filtration surgery model will be successfully built.(3)The effective componment 2 of Qing Guang An and MMC can obviously inhibit the multiplication of inoblast and the expression of type I collagen,reduce the hyperplasia of scar tissue,which has obvious effect for preventing scarring of filtration canal.(4)It suggests that orally taking the effective componment 2 of Qing Guang An does not has obvious side effects,which is much more safe than MMC,and may be a new and safe medicine to preventing scarring of filtration canal after glaucoma surgery and thus improve the success rate of the surgery.
