The current study shows that lipid peroxidation is one of the most important reasons for senescence,cancer,cardiovascular and other diseases.The antioxidant activity of plant polysaccharides have some correlation with the anti-tumor activity,prevention and treatment of many diseases.Therefore,screening of the natural polysaccharide antioxidant has attracted more attention.With the natural products polysaccharide as the research object.Study on the analysis and preparation of the polysaccharides respectively,the separation and antioxidant activity of the pine cone polysaccharides and Codonopsis Compound polysaccharide were studied,the preparation and antioxidant activity of Codonopsis Compound polysaccharide granules.The main contents are as follows:1.Made a significant improvement on the method for content determination of polysaccharides--DNS method.It have received multiple good effect and practicability.DNS method is one of the methods for the content determination of polysaccharide and has many outstanding advantages.When calculating the polysaccharide content,it needs to take the hydrolyzed polysaccharide coloured solution measured value minus the unhydrolyzed polysaccharide coloured solution measured value.This.difference calculating method is calculating the content of polysaccharide at the same time,also can remove the impurity interference.Its main deficiency is increased the corresponding difference calculation procedure.In the new method established in this experiment we direct use the hydrolysis after colored as the blank solution of colorimetric determination and replacing the original blank solution.The calculated process by hydrolysis determination minus unhydrolysis determination was omit and the multi-step operation corresponding was also omit at the same time.The results in testing by two methods were very close and methodology method met the requirements.The improved DNS method can simplify the calculation and operation.It also can avoid the absorption interference on the determination results by using chromogenic reagent as the blank solution and have multiple good effect.This method can be used for the determination of polysaccharides in pine cone and provide reference for other polysaccharide determination.2.Establish a new method for the determination of the Compound Codonopsis polysaccharide granules-amylase method combined with improved DNS method.It overcame the interference of starch to the determination of the effective component polysaccharide and the literature has not been reported in similar work.When determining the content of the Codonopsis pilosula compound granules by the improved DNS method,we encounter difficulties.Because the starch is polysaccharide and the results is real reacting the total polysaccharide content which determined by the improved DNS method.The total polysaccharide was include the effective components of polysaccharides and starch.There was no corresponding solutions in the literature.In this experiment,we try the following ideas:First remove the starch of the granules,then converts the starch into small molecule disaccharides with the amylase method.Using the alcohol soluble difference between the polasaccharides and disaccharides,the polysaccharide was separated by alcohol precipitation method.To determine the content of the Codonopsis pilosula compound granules by the improved DNS method.It can avoid the interference of excipient starch.The results show that combined amylase method and DNS method for determination of polysaccharide in Codonopsis pilosula compound polysaccharides granules can overcome the interference of starch to the determination of the effective component polysaccharide.The result of methodology determination met the requirements.This method can be used for the determination of polysaccharides in codonopsis pilosula compound polysaccharides granules and provide reference for other polysaccharide determination in Chinese medicine preparation with starch as excipient.3.To prepare different molecular weight Pinus koraiensis pine cone polysaccharide,study the antioxidant activities of different molecular weight polysaccharide,and explore preliminary the relationship between polysaccharide purification and its activity.In this experiment the different molecular weight polysaccharide of Pinus koraiensis pine cone were prepared by using membrane separation technology and purified further by removing the protein.The antioxidant activities of all polysaccharide products were investigated using various assays in vitro,including DPPH radicals scavenging ability and reducing power.Results showed that all the polysaccharides produced by different methods were found to exhibit antioxidant activity in vitro.The concentration of polysaccharide and its activity was positively correlated.The antioxidant activity of polysaccharides purified by deproteinization became weaker.It deserves attention that when preparing active polysaccharide,proteins and pigments shouldn't be removed blindly and a corresponding active experiment is often necessary.4.Based on the above research activity of single plant polysaccharide and the activity of Chinese herbal medicine compound polysaccharide was studied.To study the antioxidant activities of Codonopsis pilosula compound polysaccharides prepared by different methods,explore the relationship between polysaccharide purification and its activity.Codonopsis pilosula compound polysaccharides were isolated by membrane separation,purified further by removing the protein and pigment.The antioxidant activities of all polysaccharide products were investigated using various assays in vitro,including reducing power and scavenging ability on hydroxyl radicals,DPPH radicals,superoxide radicals.All the polysaccharides prepared by different methods showed antioxidant activity in vitro and the activity of polysaccharide increased with the increasing concentration.The antioxidant activity of total polysaccharides became weaker after purifing by deproteinization and decoloration.The Codonopsis pilosula compound polysaccharides had better antioxidant activity in vitro-and the above experiment conclution was further verified.When preparing active polysaccharide,proteins and pigments shouldn't be removed blindly.When preparating active polysaccharides it should pay attention to blindly follow the inertia of thinking.It is necessary to provide activity experiment for reference.5.The Codonopsis pilosula polysaccharide compound was separated to different molecular weight polysaccharide and the antioxidant activity of these polysaccharide were studied respectively.In the experiment the different molecular weight polysaccharide were prepared using ultrafiltration membranes.The antioxidant activities of all polysaccharide products were investigated using various assays in vitro,including reducing power and scavenging ability on hydroxyl radicals,DPPH radicals,superoxide radicals.All the different molecular weight Codonopsis pilosula compound polysaccharides showed antioxidant activity in vitro and good dose-effect relationships.This experiment found that the antioxidant activity of Compound Codonopsis polysaccharide were increased with its molecular weight.6.This experiment first studied the the molding process,quality inspection and the antioxidant activity of Codonopsis pilosula compound polysaccharide granules.In the forming process,optimizing the proportion of the granuals' excipient species and raw material by the contrast test.With dilute ethanol as the wetting agent for wet legal system grain,drying,granuletion.Codonopsis pilosula compound polysaccharides granules showed good antioxidant activity in vitro and had reducing power and scavenging ability on hydroxyl radicals,DPPH radicals,superoxide radicals.The concentration of the samples was positively correlated with its activity.Therefore,the Compound Codonopsis polysaccharide also exhibit good antioxidant activity when it was prepared into granules.This experimental results can provide basis reference for the subsequent development of the Codonopsis Compound polysaccharide preparations.This paper established two new analysis methods,prepared and separated of pine cone polysaccharide and Codonopsis Compound polysaccharide.Studied the antioxidant activity of the pine cone polysaccharide and Codonopsis Compound polysaccharide granules.On the above series of studies,there had been made progressed and there is no literature have the same work report.It deserves attention that the pine nut shell polysaccharide from waste resources.It is necessary to further study and will have a very good development prospect.In the research field of natural polysaccharides,there are more researches to the activity.There are less researches to the correlation between the degree of purification and activity.There are many researches to the single herb polysaccharide and less researches to the compound Chinese herbal polysaccharide.The relationship between
