Lycorine was found to have anti-tumor effects,has been the research focus at home and abroad.It is an isoquinoline alkaloids which is rich in Amaryllidaceae plants for example crinum and Lycoris.In vitro experiments It can inhibit the growth of many tumor cells,such as breast cancer,colon cancer,nasopharyngeal throat cancer and so on.The anti-cancer effects of Lycorine can be divided into two blocks,inhibit DNA and protein synthesis and induction apoptosis.Many studies have shown that it can increase the concentration of Ca2+.The endoplasmic reticulum is the main storage organelles for Ca2+.Change the homeostasis of Ca2+ in the Endoplasmic reticulum and protein accumulation of unfolded or misfolded can lead to endoplasmic reticulum stress response.This article from the endoplasmic reticulum stress and endoplasmic reticulum protein expression of apoptosis-related perspective to explore the molecular mechanism of Lycorine anti-breast cancer.Western Blot was used to check the effect of Lycorine on Bip/GRP78?XBP-1?caspase-12?chop protein expression in MCF-7 cells.Experimental results show that different dosages of Lycorine could effectively induce apoptosis in MCF-7 cells after 48 hours,and dose-dependent manner.Lycorine can up-regulate the expression of Bip/GRP78 in a dose-dependent manner,compared with the control group the percentage value is(109.39±0.57)%?(119.62±0.29)%?(159.5±0.31)%,compared with the control group was significantly higher(p<0.01);Lycorine can up-regulate the expression of XBP-1 in a dose-dependent manner,compared with the control group the percentage value is(101.62±0.15)%?(122.9±0.48)%?(130.59±0.69)%,compared with the control group was significantly higher(p<0.01);Lycorine can up-regulate the expression of caspase-12.compared with the control group the percentage value is(101.38±0.48)%?(131.33±0.54)%?(151.77±0.73)%,compared with the control group was significantly higher(p<0.01);Lycorine can up-regulate the expression of CHOP/GADD153 in a dose-dependent manner,compared with the control group the percentage value is(100.72±0.1)%?(103.92±0.7)%?(108.11±0.44)%,compared with the control group was significantly higher(p<0.05).Summarily,Lycorine allows MCF-7 cells endoplasmic reticulum stress and protein expression by upregulating Bip/GRP78 to alleviate endoplasmic reticulum stress.The endoplasmic reticulum stress reaches the high level,XBP-1 expression level was significantly increased apoptotic cells into irreversible process,and ultimately through the expression of caspase-12 and CHOP/GADD153 proteins,,by which induce apoptosis of MCF-7 cells through the endoplasmic reticulum pathway.
