Objective Establish methods of inducing deficiency-heat syndrome in MRL/lpr mices by hot Chinese medicine and evaluate the model.To evalutate and explore the effect and mechanism of Artemisia apiacea-turtle shell herb-pair with respect to treating MRL/lpr mices deficiency-heat syndrome,providing an experimental basis for clinical applications.Methods The establishment and evaluation of deficiency-heat syndrome model in rats:decoction of three Chinese medicines with thermal property-aconite,cinnamon,dried ginger were administered intragastrically to rats for four weeks,the changes of general condition,body weight,temperature,water intake,food intake and various blood biochemical indexes of rats were examined for evaluating deficiency-heat syndrome model.Pharmacodynamic study of Artemisia apiacea and Turtle shell herb-pair to deficiency-heat syndrome model:select the rats with deficiency-heat syndrome which were divided into seven groups,deficiency-heat syndrome group(DHS),Artemisia apiacea group(AA),Turtle shell group(TUR),Herb-pair(HP),positive control group(ZB),and set blank control group(NC),the rats were given corresponding medicines for four weeks.According to the objective indicators of deficiency-heat syndrome and the blood biochemical indexes of MRL/lpr mice to research the compatibility mechanisms of herb-pair.Pharmacokinetic study of artemisinin:establish a HPLC-MS method for the determination of artemisinin in MRL/lpr mice plasma.PKSolver pharmacokinetic Software was used to calculate pharmacokinetic parameters and the compartm-entalmodel.Results The establishment and evaluation of deficiency-heat syndrome model in MRL/lpr mices:mices in model group looked abnormal agitated,water intake and food intake increased,compared with the normal control group,slow weight gain and temperature had a trend of decreasing,T3?T4 and E2/T increased significantly,the content of COR had the tendency which elevated.Pharmacodynamic study of Artemisia apiacea and Turtle shell herb-pair to deficiency-heat syndrome model:Compared to the blank control group,MRL/lpr mices in model group looked a bit manic,body weight was decreased and the difference was significant(P<0.01),T3?T4?E2?cAMP?LD?LDH levels in blood serum increased significantly,but T and cGMP reduced obviously(P<0.01,P<0.05).Compared to the model group,body weight had different degrees of increasing in treatment groups,water intake and food intake decreased,T3?T4?E2?COR?cAMP?LD levels in blood serum decreased,T and cGMP increased after the two Chinese medicines combined,it also can significantly reduce the adrenal index of deficiency-heat MRL/lpr mices(P<0.01).High dose group had the same efficacy to the positive control group,no significant difference.The pharmacokinetics study of artemisinin in deficiency heat syndrome rates:Pharmacokinetic study of artemisinin:A HPLC-MS method for the determination of artemisinin in rat plasma was established.The mobile phase consisted of acetonitrile and ammonium acetate of water(70:30,v/v),the flow rate was set at 0.2mL/min,UHPLC XB-C18(2.1mm ×150mm,1.8?m)chromatographic column as analytical column,ESI was applied,Multiple reaction detection method for positive ion detection.The ionic detection for quantitative analasis were m/z305.The linear calibration curves were obtained in the concentration range of 8.48?25.44?g/mL.The lowest limit of quantification was 6.71?g/mL.The recovery was96.83%Conclusions This passage illustrate the successful to Establish methods of deficiency-heat syndrome in MRL/lpr mices by hot Chinese medicine and evaluate the model.By discussing the Deficiency-heat syndrome on multiple aspects such as endocrinium,immunology,metabolism,pathology and hematology,as well as analyzing the indexes of the syndrome,various causes of the syndrome were exposed,which laid the foundation for further studies.This essay focuses on Artemisia apiacea-turtle shell herb-pair.The different effects between isolated medical application and combined application of the herb-pair to the syndrome was analyzed by observing MRL/lpr mices general situation,body weight,body temperature,water intake,food intake and kinds of blood biochemical indices.Then the material basis and the mechanism of the herb-pair before and after its combination were exposed.Method of plasma pretreatment by UHPLC-MS was built.Method of measuring Artemisia apiacea content within the body of the gavage group rats was established.To research the pharmacokinectics of artemisinin which of artemisia apiacea and turtle shell herb-pair can provide the basis for compatibility of herb-pairs.
