Objective:To elucidate the chemical components in 2M and its analogous and the absorbed components after oral administration of 2M and its analogous,it provides a method for analyzing the constituents of Chinese medical formula in vitro and in vivo,revealing its direct role material in the body and helping to explore the effective substance of 2M and its analogous.lt will be meaningful to guide clinical medication rationally and improve its quality standards so as to enhance the competitiveness of drugs.Methods;According to the theory of Serum Pharmacochemistry of TCM,UPLC-MS/MS technique was applied to detect and characterize the constituents of 2M and its analogous and the absorbed components and their metabolites in rat plasma after oral administration of 2M and its analogous.The quality of 2M and its analogous also will be assessed objectively.Results:1.Analysis of the constituents in vitroA systemic analysis method of 2M and its analogous constituents in vitro was established.An injection run of the 2M and its analogous sample was finished in only 24min,and excellent chromatographic separation and sensitive detection were obtained.By the intuitional inspection to the UPLC-MS/MS chromatograms,44 compounds were detected and identified in 2M,of which 24 compounds were detected in positive ion detection mode and 20 compounds were detected in negative ion detection mode;44 compounds were detected and identified in 3M,of which 25 compounds were detected in positive ion detection mode and 19 compounds were detected in negative ion detection mode;50 compounds were detected and identified in 4M,of which 30 compounds were detected in positive ion detection mode and 20 compounds were detected in negative ion detection mode;Detected chemical composition mainly including:Phellodendrine?magnoflorine?dauricine?Tetrahydropalmatine?Jatrorrhizine?Palmatine?Berberine?obaculactone?Obacunone?3-O-Feruloylquinic acid?Sanleng acid?Tetramethyl-O-scutellarin?N-candicine?b-Glucogallin?Amurensin?2-(p-hydroxyphenyl)ethanol 1-O-?-D-glucoside and so on.which belong to Alkaloids?Organic acids?Three terpenoids?2.Analysis of the constituents in vivoBased on the theory of Serum Pharmacochemistry of TCM and UPLC-MS/MS technique,an injection run of the sample of dosed rat plasma was finished in 24.0 min.Through applications of various data processing technology,19 compounds were detected and characterized in rat plasma after oral administration of 2M;18 compounds were detected and characterized in rat plasma after oral administration of 3M;18 compounds were detected and characterized in rat plasma after oral administration of 4M.3 ConclusionAccording to Serum Pharmacochemistry of TCM(Traditional Chinese medicine),the comprehensive application of UPLC-MS/MS technology contributes to the global analysis of the constituents of SGT in vitro and in vivo.The results help to classify the material basis responsible for the therapeutic effects of 2M and its analogous?Some potentially efficient markers have been identidied according to the characteristics of compounds found in dosed serum,which are posible to serve as eriteria in assessing the quality of 2M and its analogous.The method which combines analyzing the constituents of TCM in vitro and in vivo and identify efficient markers baseing on the behaviors of compounds in vivo will be more significant in revealing the intrinsic quality of TCM and the effective substance of TCM.
