Macfadyena unguis-cati(L)A.Gentry(Bignoniaceae)is an ornamental climbing plant,widespread in Lesser Antilles,Mexico and Egypt.It is native to tropical America and introduced into China as ornamental plant in early time.Macfadyena unguis-cati is used in folk medicine to treat snakebite,dysentery,inflammation,stomach bloated,and rheumatism disease.Also,there are reports on its use in the treatment of venereal disease and as a quinine substitute for malaria.Now,it was found that M.unguis-cati shows obvious effect on anti-inflammatory,anti-oxidation,anti-tumor and antibacterial activity.First,M.unguis-cati was extracted by ethanol,followed by extracting of solvent with different polarity,and then the extracts were tested for their effect on anti-tumor and antioxidant activity.Chloroform portion and fraction D give a strong inhibitory effect on proliferation of human hepatocellular carcinoma cells SMMC7721 with IC50 value 53.5±28.61 and 52.17±19.03 ?g/mL respectively;FrE,FrF,FrG and FrH inhibited proliferation of SMMC7721 significantly,with IC50 value 27.64 ±14.31,51.88 ± 22.21,35.94±1.31 and 22.89 ± 4.79 ?g/mL respectively.N-butanol portion share a similar antioxidant capacity with Vc at 0.8 mg/mL.Also,n-butanol portion had excellent radical scavenging activities,with IC50 value 5.83±0.29 ?g/mL.Radical(DPPH)scavenging activity of portion A from chloroform had increased significantly,with the IC50 value 3.95± 1.8 ?g/mL.In a word,chloroform portion of M.unguis-cati show good effect on anti-tumor activity and portion A purified by RP-18 show good effect on antioxidation activity,basing on anti-tumor and antioxidant activity screening model.Secondly,seven compounds were isolated by the methods of silical gel,sephadex LH-20,and reverse-phase silica gel column.Six compounds,including decaffeoyl acteoside(ML-1),lyoniresinol-3a-O-?-D-Glucopyranside(ML-2),cirsimarin(ML-3),cirsimaritin(ML-4),ursolic acid(ML-6)and 1,2-Benzenedicarboxylic acid,1,2-dibutyl ester(ML-7),were elucidated on the basis of chemical and spectral methods.Compound ML-1,ML-2,ML-3,ML-4 and ML-6 were first isolated from M.unguis-cati.At last,anti-tumor activity of compounds were detected by the MTT method,it was found that compounds ML-4 inhibited the growth of SGC7901,Bel7402 and Chang-liver with the IC50 value 5.89±2.52,73.65±7.53 and 71.24± 10.99 ?g/mL respectively.No obvious anti-tumor activity was found on the growth of SMMC7721 and HepG2;ML-6 show a inhibitory effect on SMMC7721,HepG2,SGC7901,Be17402 and Chang-liver with IC50 value 29.71±2.70,68.26±14.60,22.13±1.35,33.41±0.14 and 41.49±2.17 ?g/mL respectively;ML-1,ML-2 and ML-3 show few anti-tumor activity on all the cells.According to the results,ursolic acid as anti-cancer active ingredients of M.unguis-cati was confirmed.
