| Objective:through animal experiment observation acupuncture point pretreatment of paroxysmal atrial fibrillation in the rat heart bipolar parameters between QTc period and the changes of the ultrastructure of rat heart muscle,heart,and with other interference factors on paroxysmal atrial fibrillation in the rat of QTc interphase and ultrastructural changes of rats,to explore the effectiveness of the closed cavity pretreatment in the treatment of paroxysmal atrial fibrillation in the acupuncture,for clinical use of this method provides the theoretical foundation treatment of paroxysmal atrial.Methods:1.The selection of weight 250-300-g male,healthy,clean level of Wistar rats,after anesthesia in rats by tail vein injection of acetylcholine-calcium chloride mixture for seven consecutive days to establish animal model of paroxysmal atrial fibrillation in rats.The rats were randomly divided into normal control group and drug group,acupuncture preconditioning group and model group.Each group rats processing methods:acupuncture preconditioning group:the rats closed in on both sides acupuncture point acupuncture needle after half an hour,parallel injection mold drugs,once a day,7 days;Drug groups:made mold injected rats with drugs once a day,2 times daily irrigation suit is equivalent to 4.5 mg/(kg·d)statement of amiodarone,a total of seven days;Model group,model group rats tail intravenous building drugs once a day,a total of seven days;Normal groups:normal group of rats after anesthesia,caudal vein injection(1 mg/kg)of physiological saline,a total of 7 days.On the eighth day of materials and related indicators of detection.Each group atrial ultrastructure of rats and the change of atrial structure observation:By HE staining and observed under electron microscope atrial muscle tissue of rats,and to compare each rat heart changes of the ultrastructure and organization structure.Through the calculation of QTc interphase of rat,to compare between groups in the rat heart after a parameter of QTc interphase values,as the outcome indicators.Statistical analysis:the homogeneity of variance analysis was carried out on the data and normal inspection,all data using spss17.0 statistical software package processing,data with mean standard deviation,according to comparison between groups by using single factor analysis of variance and t test,with P<0.05,with statistical significance.Results:1.The rat made mould injection drug immediately after anesthesia in typical paroxysmal af ecg manifestations(p wave disappeared,replaced by irregular f wave,r,r-r interphase),to sinus rhythm,f wave,p wave appears to atrial fibrillation.The model group rats in injection mould drug maker 1 day,4 days,7 days QTc interphase has the remarkable difference compared with normal group(p<0.01).3.Injection drug 7 days later,acupuncture preconditioning group and drug group of QTc interphase compared with model group of QTc interphase significantly shortened(p<0.05).4.The period between drugs and acupuncture preconditioning group of QTc built in injection mold drug 7 days compared with no significant difference(p>0.05).5.HE staining results:paroxysmal atrial fibrillation group showed irregular arrangement of myocardial cell and the nucleus sizes and intracellular muscle fiber fracture,fracture and part is focal myocardial fibers have muscle fiber fusion;The drug therapy group cell interstitial edema,slightly slightly and tidy arrangement of myocardial cell;Myocardial cell structure is complete,acupuncture preconditioning group arrangement not entire,slightly slightly clear nucleus,fiber cell shape rules,mild edema,myocardial fibers occasional owe rules.In histological study above,acupuncture preconditioning group and drug group is better than that of model group.Conclusion:1,the rat tail vein injection of acetylcholine-calcium chloride mixture paroxysmal atrial fibrillation in 7 days,in the electrophysiological changes of electrocardiogram(ecg)and heart and the occurrence of clinical atrial fibrillation,building success,is reproducible,can be used in the study of basic experiment.2,acupuncture in den pretreatment in improve paroxysmal atrial fibrillation in the rat ventricular bipolar parameters of QTc interphase on slightly better than drug group,has significant difference with model group,the rats in addition to the polar and bipolar process returns to normal,reduce the cutting-edge reverse type of ventricular tachycardia and reduce the mortality of rats.3,acupuncture preconditioning group can reduce the paroxysmal atrial fibrillation in the rat heart muscle fibers,cell morphology,such as mitochondria damage degree,similar to the drug group,protect the paroxysmal ultrastructure of rat heart. |
