Objective By observing the influence of HIF-1 alpha(Hypoxia-Inducible Factor-1 alpha,alpha HIF-1)expression in Hypoxic Ischemic Brain Damage(HIBD)neonatal rats which were treated by Tanshinone ?a(TSA),we initially explored the protective effect of TSA for the newborn rat brains.Methods Sixty 7-days SD rats were randomly divided into 5 groups in the experiment:sham-operated(Sham group)12 rats,model group 12 rats and Tanshinone ?a treatment groups(TS A Groups)36 rats.The TS A Groups were divided into high-dose group(short for TSA-HD Group,16mg/100g,12 rats),middle-dose group(TSA-MD Group,8mg/100g,12 rats)and low-dose group(TSA-LD Group,4mg/100g,12 rats).In the Sham group,the left common carotid artery(LCCA)were separated,but not ligatured.And in all the other groups,the RICE method was employed to make HIBD model of newborn SD rats.Behavioural indicators are used to observe behavior changes among the newborn rats after model-building,and with FQ-PCR method,the quantity of.HIF-1? gene expression was determined in every groups.HE dyeing method was employed to observe brain tissue pathological changes,with HIF-1? immunohistochemical markers dyeing in situ,positive expression were compared under microscope.With SPSS 19.0 statistical software,Tamhane's T2 test was employed for statistic analysis.When the probability was less than 0.05,two groups were considered as statistically different.Results 1.The model-making of HIBD was successful,which was proved by abnormal behavior occurred among the HIBD rats and ischemic pathological changes happened in the HIBD rat brains.2.It is shown by HE stain that TSA can improve the hypoxia-ischemic damage in brain tissue,RT-PCR and immunohistochemical markers results manifested that TSA can up-regulate the expression of HIF-1? gene in brain cells.When the expression of HIF-1? was higher,the pathological changes was better.3.Among the TSA Groups,HIF-1?expressed most in TSA-HD Group,followed by TSA-MD Group and TSA-LDGroup(TS A-MD Group more than TS A-LD Group).Conclusions 1.After the HIBD rats was operated,rats behaved abnormally.Shown b y HE stains,there were ischemic pathological changes in the HIBD rat brains,all of above proved that the building of HIBD model was successful.2.According to HE st ains,immunohistochemical markers and RT-PCR results,TSA can provide brain tissue with a protection effect against hypoxia-ischemic damage,and the protection effect agg randized as the increasing of HIF-1? gene expression.TSA perhaps took effect by up-regulating the expression of HIF-la gene.3.As TSA dose increased,the expression o f HIF-1? gene increased,it meant that improving the TSA dose within a certant limits,the expression of HIF-1? can be improved.
