Objective:To investigate the effects of Maixiansan on MUC2 and CDX2 in colon of the rats with ulcerative colitis and explore the therapeutic effect and mechanism of Maixiansan of ulcerative colitis,provide a theoretical basis for the Maixiansan in clinical treatment of ulcerative colitis.Methods:40 SD male rats were randomly divided into normal group,model group,mesalazine slow release particles dose group,Maixiansan group,in addition to the normal group,other groups were treated with 5%dextran sodium sulfate(DSS)in their drinking water for 7 days to induce ulcerative colitis.After successful modeling,each experimental group was administrated NS,mesalazine,and Maixiansan respectively.At the same time,observe the general condition,weight change,action,stool,blood in the stool of each rat.Two weeks later,all survival rats were sacrificed.The colon tissue was taken to do biopsy and pathology.The expression of MUC2 and CDX2 in colon tissue were detected by Immunohistochemical technique.Results:1.All the rats in the DSS model group can be observed the changes of the symptoms such as intermittent diarrhea with mucus or blood at stool.Oedema,hyperemia and anabrosis were observed in the colonic mucosa.Combined with the consequence of pathological section,we were sure that UC model of rats was made successfully.2.Mesalazine slow release particles and maixiansan notably reduced macroscopical scores of colon tissues obviously(P<0.01),Compared with Mesalazine slow release particles group,maixiansan group had statistically decrease(P<0.05).3.The content of mucin MUC2 was notably decreased in model group compared with normal group(P<0.01),while they were increased in mesalazine slow release particles dose group and Maixiansan group,especially in Maixiansan group(P<0.05).4.The content of mucin CDX2 expressed obviously in normal group and had a comparatively lower expression in model group(P<0.01).Then,we also discovered CDX2 expessed more relatively in mesalazine slow release particles dose group and Maixiansan group compared with model group.Moreover,this difference was more significant in Maixiansan group(P<0.01).Conclusion:Maixiansan could notably improve general condition,reduce macroscopical scores of colon tissues and promote anabrosis healing,and its potential mechanism may be related to up-regulation of MUC2 and CDX2 in colonic mucosa.
