Objective:To establish determination method of main component in Guhong injection,and establish HPLC fingerprint of Guhong injection,provide reference for internal quality control of Guhong Injection.Methods:(1)Using AlltimaTM C18 chromatographic column(4.6 mm*250 mm,5 microns),acetonitrile-0.1%formic acid solution as mobile phase gradient elution,flow rate of 0.9mL min-1,detection wavelength of 210 nm,260 nm,403 nm.(2)Using the Folin-Ciocalteu colorimetric method,according to the consistent degree of the maximum absorption wavelengths of hydroxysafflor yellow A and kaempferol relative to Guhong Injection,we selected the appropriate reference and color reaction condition to content determination.(3)The application of HPLC,using AlltimaTM C18 chromatographic(250 mm*4.6 mm,5 microns),0.1%formic acid aqueous solution-acetonitrile as mobile phase,gradient elution,in 0 to 7 min,detection wavelength is 239 nm,switch to 260 nm after 5 minutes,switch to 280 nm after 36 minutes,establish Guhong injection fingerprint.Results:(1)Acetyll-glutamine,uridine,adenosine,guanosine,lilacsglycosides,hydroxy safflower yellow pigment A,dehydration of safflower yellow pigment B,their linear relation is favorable in the range of test(r>0.9996),precision test RSD<2%,stability test with in 24 h RSD<2.3%,the average sample recovery rate was 99.12%?100.68%,RSD<2%.(2)In hydroxy safflower yellow pigment A,total polyphenol content in 10 batches of GuGong injection were 9.94,9.55,9.75,9.67,9.84,10.03,9.81,9.52,9.88,10.09 mg mL-1.Average sample recovery rate of total polyphenol was 98.11%,RSD was 1.68%(n=6).(3)All ingredients of Guhong injection get a better separation,with hydroxy safflower yellow pigment A as the reference peak,identified 21 common peak,fingerprint similarity of 10 batches of Guhong injection is greater than 0.973.Conclusion:(1)The established HPLC-DAD method is simple and reliable,and can simultaneously determine Guhong injection in a variety of active ingredients,can be used for quality analysis of the preparation.(2)The Folin Ciocalteu-method is simple,accurate,high sensitivity,is a reliable method of quantifying the total polyphenols in Guhong injection.(3)HPLC method can separate each element in the Guhong injection at the same time,the method is simple and accurate.stable and reliable,and can be used as an establishment method of Guhong injection' s fingerprint.
