Glioma is one of the most common intracranial tumors,which has a high recurrence rate and high mortality.The 5-year survival rate of glioblastoma is only 5%.So far,the etiology and pathogenesis of glioma is still unknown.Generally,genetics,gene mutation and physical and chemical factors are association with development of glioma.In recent years,A number of articles have reported that they have detected nucleic and protein of human herpesvirus 6(HHV-6)in glioma tissue.Our previous isolated HHV-6 from the fluid specimens from giloma cysts and dectected HHV-6 infection was 42.5%(17/40)in glioma tissue,significant different from normal brain.We speculated that HHV-6 infection play a role in the occurrence and development of glioma.Study bioinformatics of HHV-6 DR7 gene is potentially transforming gene and has trans-activation activity.This study was to investigate the effect of HHV-6 DR7 gene expression on biological characteristics of human glioma U87 cells.This paper was divided into tow parts:In the first part,Gene was amplified from the plasmid carrying the DR7 gene of HHV-6 by PCR,lentiviral vectors pLenti6.3-DR7-IRES2-EGFP was constructed and packed using 293T cells,the virus stock solution was concentrated,and the virus titer was determined.HHV-6 DR7 gene recombinant lentiviral infection U87.A separate U87 cell line stably expressing DR7 was established after flow cytometry and blasticidin screening.This was the foundation for the follow-up study of DR7 gene on the biological characteristics of U87.In the second part,we used CCK-8,soft agar cloning,cell scratch,transwell and chick chorioallantoic capsule experimental to investigate the effect of HHV-6 DR7 gene on cell proliferation,migration,invasion and angiogenesis of human glioma U87.The results showed that DR7 gene could promot U87 cells in cell proliferation,migration,invasion and angiogenesis.Cell cycle analysis showed that DR7 gene can promote cell cycle runs,the proportion of S phase and G2/M phase were significantly increased.In summary,this experiment was successfully constructed stably expressing HHV-6 DR7 gene U87-DR7-EGFP cells.the DR7 gene can promote U87 cell proliferation,migration,invasion and angiogenesis.This study provides new ideas and targets to treat human glioma.
