Studies On The Quality Standards Of Selaginella Moellendorfii Hieron.Extract And Its Action Of Anti-Gout

Gout is a common disease.At present,the main medicine used to treat gout is colchicine,etc.,but it has side-effects,and it cannot avoid recurrent attacks.Therefore,it makes significant sense to research and develop new drugs which is safe and effective to gout.Selaginella moellendorfii Hieron,which is from Pteridophyte Selaginellaceae,is a frequently-used Chinese herbal medicine in folk.It can clear heat,expel miasma,cure cancer and stop bleeding,acute maladies such as cancer,pneumonia,tonsillitis,conjunctivitis,and mastitis.There are studies which have shown that biflavonoids of the plant can inhibit xanthine oxidase activity,and can lower the blood's uric acid level effectively to relieve pain caused by gout.Consequently,researches on the quality standards of its extraction and its action of anti-gout,provide a favorable basis for the development of new medicine used to cure gout.This project has set quality control specification to the extractive of Selaginella moellendorfii Hieron,and has built the foundation for related new drug through initial study on anti-gout.The main subject of research and results are as follows:1.Selaginella moellendorfii Hieron.extract quality standardsTLC was used for the identification of the extract with the amentoflavone as reference substance,and the results showed that the method of TLC had fine separability and excellent durability.HPLC was adopted to perform a quantitative research on the contents of the amentoflavone of the extract.The results of the linear relationship,precision,stability and reproducible were good,and the recovery rate was qualified.Three batches of extracts were measured,the level of the contents of amentoflavone should not less than 1.2mg per 1 mL in the solution of Selaginella moellendorfii Hieron.extract.The level of the contents of amentoflavone should not less than 0.12mg per 1 mg in the dry extract of Selaginella moellendorfii Hieron.extract.2.Acute toxicity testMice stomachs were irrigated with the maximum concentration and the maximum volume drug of Selaginella moellendorfii Hieron.extract one time(corresponding to 40 times of human clinical equivalent dosage),and were observed for one week,no immediate abnormalities were found.3.Study on the anti-gout effect of the Selaginella moellendorfii Hieron.extract.The anti-gout effect of the Selaginella moellendorfii Hieron.extract was studied,and results compound the Selaginella moellendorfii Hieron.extract could significantly inhibit the sodium urate crystals-induced toe swelling of rats,hyperuricemia and TNF-? of mice.The treatment group was significantly different compared with the model group,what's more,it was not significantly different compared with the positive group.In summary,the above studies showed that the standard is controllable,the effect of anti-gout is remarkable.These studies have laid a foundation for the anti-gout drug development which uses the exctract as intermediates.
