BackgroundSince the last century,lung cancer has become the leading cause of cancer death around the world,and its morbidity and mortality have rising.In China,it's also the leading cause of cancer deaths.Lung cancer is threatening human health,therefore effective treatments has become a hot topic.RNA interference(RNAi)is a gene silencing technology developed in recent years,which has high efficiency,specificity,stability and other notable features.Gene therapy requires a valid target.Ryanodine receptor 1(RyR1)is a high molecular weight tetramer calcium channel protein which plays a crucial role in the calcium signal generation and excitation-contraction of muscle cells.Research reports suggested that RyR1 genes play an important role in different kinds of tumor.This project evaluates the impact on the activity of the cell biology of lung cancer,whose RyR1 gene is silencing.In order to find a new lung cancer metastasis-related genes,we should provide new molecular targets and the mark of recurrence and metastasis of lung cancer treatment and prognosis.ObjectiveTo investigate changes in biologic properties of non-small-cell lung cancer(NSCLC)A549 cells whose Ryanodine receptor 1(RyR1)expression was suppressed by short interference RNA(siRNA).Methods1.siRNA and negative control segment were designed and synthesized to transfect lung cancer line A549.2.RT-PCR were used to determine mRNA expressions of RyR1 before and after transfection.3.MTT assay was used to measure the changes of cell apoptosis.4.Cellular cycle and apoptosis were detected with flow cytometry.Results1.The expression of RyRl gene decreased after transfection as was shown by RT-PCR.2.With MTT assay,we detected a decline of cellular activity.3.There was a significant rise in the percentage of G0/G1-phase cells and apoptosis rate compared with the blank group(normally-cultured A549 cells)(P<0.05)and control group(negative control fragment)(P<0.05)separately.ConclusionThe RyR1 gene silencing slows the growth rate of lung cancer cells.With more cells in G0/G1-phase,the expression of silent RyR1 gene could both promote the apoptosis rate of lung cancer cells and suppress the cellular growth.Thus RyR1 gene could serve as a new strategy for the genetic treatment of NSCLC.
