| REGγ protein belongs to the 11S proteasome activator,it can activate proteinsome,to de,grade some target proteins,including p21,p16,SRC-:3,and so on.RFGγ also plays a crucial role in tumorigenesis and development,The abnormally high expression of REGγis found in a large scale of tumor tisues.There.i s also high expression of REGγ in thyroid tumors,especially in poorly-di fferentiated thyroid carcinoma.This paper was aimed to study the role of REGγ in thyroid tumors.We study the role of REGγ by focusing on its effect on thyroid carcinoma differentiat ion state,the migration abili ty of thyroid tumor cells,cell proliferal ion and cell apoptosis,and we explore the molecular mechanism.Conclusions were drawed that:REGγ inhibited the expression of thyroid-specific genes,REGγpromoted the migration ability of thyroid tumor cells,cell proli feration was promoted by REGγ,REGγrendered cel Is anti-apoptosis effect,REGγ helped PKAαtranslocated into nuclear much easier,REGγ decreased the protein stability of PKA a and it inhibited t he expression of PKAα.The regulation of thyroid-specific gene expression by REGγ was mediated by the cAMP-PKA signal pathway.Besidos,both REGγand MAPK signal pathway were involved in the regulation of thyroid-specific gene expression.It seemed REGγ played a crucial role in tumorigenesis and development,so studying on role and mechanism of REGγ had critical clinical significance.REGγinhibited the expression of thyroid-specific genes,including NIS gene,which made the clinical treatment with radioactive I131 have litile effect.REGγ rendered cells anti-apoptosis ability,which also made the treatment difficult.We can regard REGγ as a target protein in future study and clinical treatment by decreasing its expression or lowering its activi ty,which would have positive meaning in thyroid tumor treatment. |
