| BackgroundEpigenetics refers to does not involve DNA sequence of gene or protein expression changes,only changes in gene expression and function,and can produce genetic phenotypes.Epigenetics as a classic genetics further supplement and perfection,involved when,where and what kind of ways to apply and use the concept of genetic information.We all know that the genome contains two types of genetic information:such as sequences of DNA genetic information and epigenetics genetic information.The two kinds of information interaction,mutual constraints,and mutual balance maintain the normal function of the human body.Therefore,any kind of expression of these two factors imbalances,have possibility cause disease.The study of epigenetics,therefore,is in the field of life science in a new universal and important part of current study results show that not only of genes expression,regulation and genetic epigenetics plays an important role,and in the occurrence of tumor,aging,inflammation,regenerative medicine,angiogenesis and immune diseases such as developing and its important role.Histone acetylation enzyme inhibitors,referred to as HDACIs,can interfere with histone acetylation enzyme function,thereby increasing intracellular histone acetylation level,improving p21 gene expression level,thereby inhibiting tumor cell proliferation and induce cell differentiation and/or apoptosis.Histones to acetylation enzyme inhibitor has become a new hotspot in the research of the tumor target therapy,the inhibition of tumor cell migration,invasion,metastasis and anti-tumor angiogenesis effect have also been confirmed.Senile dementia(Alzheimer’s diseases,AD)is a progressive neurodegenerative disease,amyloid beta(beta-amyloid peptide,A β)deposition of neurotoxicity theory and brain aging is two of the most important hypotheses of Alzheimer’s disease.And Aβ is the beta amyloid precursor protein hydrolysate(β-APP).Previous findings in this research suggests that the tumor treatment has some effect of HDAC1 inhibitors on the metabolism of APP,we think that on the basis of the epigenetic HDAC1 inhibitors for the treatment of alzheimer’s may have certain effect.For an in-depth study has important significance.ObjectiveScreening several effective monomer in south medicine of traditional medicine,such as bajijiasu,protocatechuic acid,patchouli alcohol,quercetin,curcumenol.At first select the monomer can inhibit tumor cells growth and has effect on promoting apoptosis of tumor cells.Next test whether it has the function of HDAC1 inhibition,finally to determine its effect on metabolism of APP.For HDAC1 inhibitors might have a great effect for the treatment of AD.MethodsAt first test,determined by MTT method in each monomer of south medicine such as bajijiasu,protocatechuic acid,patchouli alcohol,quercetin,curcumenol work on two strains of tumer cells,N2A and SY5Y,screening to determine has inhibitory effect on tumor cell growth proliferation of acting time and the best effective concentration of monomer.Western blot method first check with HDAC1 inhibitory effect,and then use the same method to detect the expression of Bax and Bcl-2 regulation and determine its effects on tumor cell apoptosis.Finally still use this method to detect the expression of APP.ResultsIn the experimental set of drug effect time and drug concentration gradient,bajijiasu,protocatechuic acid,patchouli alcohol,curcumenol have no obvious inhibition of growth of N2A and SY5Y.Quercetin has obvious inhibition of growth of two strains tumer cells,at the same time has the HDAC1 inhibition.Further study found that the expression of quercetin on APP has a lower trend.ConclusionQuercetin is a potential of HDAC1 inhibitor,on inhibiting tumor cell growth,promoting apoptosis has strong function,and its effect on the expression of APP has down,which suggests that it may have a certain therapeutic effect for the AD and the specific mechanism of action and ways need further in-depth study. |
