| Objective To probe gene expression of NES1 and its gene methylation status in normal liver cells and liver cancer cell lines,and its impacts on gene expression and cell biology.Methods Detected the expression level of NES1 mRNA in HepG2 as well as L02 by RT-PCR and RT-QPCR.Detected the level of gene methylation with MSP.Then,with the treatment of 5-Aza-CdR,we used MTT to detect the survival rate,the expression level of NES1 mRNA with RT-QPCR and the level of cyclinD1,P21,P53 by western blot.Results NES1 mRNA expression in HepG2 compared with L02 was significantly reduced,and the level of HepG2’s NES1 exon 3 CpG island methylation was much higher than L02.After demethylation,NES1 mRNA expression of HepG2,the protein level of p21,p53 were up-regulated,meanwhile the survival rate and the level of CyclinD1 were decreased.Conclusions In HCC,low expression of NES1 mRNA relate to the gene exon 3 CpG island methylation.NES1 exon 3 methylation may be the molecular mechanisms of reducing the expression of NES1 mRNA.5-aza-dC inhibits the proliferation of HepG2 and then arrests cell cycle. |
